血小板衍生因子和真核起始因子4E在子宫肌瘤组织中的表达及意义

摘　　要：目的探讨子宫肌瘤组织中血小板衍生因子(PDGF)和真核起始因子4E(eIF4E)的表达情况及其临床意义。方法选取2013年4月-2016年10月该院收治的子宫肌瘤患者86例,行子宫切除术后,立即获取子宫肌瘤组织;另外,在至少距离肌瘤组织2 cm处获取周围正常组织67例。采用SP染色法检测标本中PDGF和eIF4E表达水平,分析PDGF和eIF4E表达情况与子宫肌瘤间的关系,并分析PDGF与eIF4E的相关性。结果 PDGF可以在子宫肌瘤组织和周围的正常组织中表达,主要集中在细胞质或细胞核中;eIF4E主要表达在子宫肌瘤组织的细胞质中;子宫肌瘤组织中PDGF和eIF4E的阳性表达率分别为80.23%(69/86)和83.72%(72/86),均明显高于周围正常组织的11.94%(8/67)和4.48%(3/67),差异均有统计学意义(均P<0.05);除子宫肌瘤的不同时期外,PDGF和eIF4E的阳性表达率与患者的年龄、肌瘤大小和肌瘤个数存在明显的相关性(P<0.05);PDGF和eIF4E在正常组织中的表达无相关性(r=0.324,P=0.395>0.05),而在子宫肌瘤组织中的表达呈正相关(r=0.765,P=0.016<0.05)。结论 PDGF和eIF4E在子宫肌瘤组织中高表达,并且与子宫肌瘤的相关参数具有密切联系,临床上可用于预测和诊断子宫肌瘤的发生与发展。另外两者在子宫肌瘤组织中的表达呈正相关,为临床上研发治疗子宫肌瘤的药物提供新的靶点。Objective To explore the expressions and significance of platelet-derived growth factor（PDGF） and eukaryotic initiation factor 4E（eIF4E） in uterine fibroids.Methods From April 2013 to October 2016,86 patients with uterine fibroids treated with hysterectomy in the hospital were selected,uterine fibroids samples were obtained;67 normal uterine tissue samples at least 2 cm far from uterine fibroids were selected.Immunohistochemical SP method was used to detect the expression levels of PDGF and e IF4 E.The relationships between expressions of PDGF and e IF4 E and uterine fibroids were analyzed,and the correlation between PDGF and e IF4 E was analyzed.Results PDGF expressed in uterine fibroids tissues and surrounding normal uterine tissues,which mainly concentrated in cell cytoplasm or cell nucleus.e IF4 E mainly expressed in cell cytoplasm of uterine fibroids tissues.The positive expression rates of PDGF and e IF4 E in uterine fibroids were 80.23%（69/86） and 83.72%（72/86）,respectively,which were statistically significantly higher than those in surrounding normal uterine tissues [11.94%（8/67） and 4.48%（3/67） ]（all P〈0.05）;except different stages of uterine fibroids,the positive expression rates of PDGF and e IF4 E in uterine fibroids were significantly correlated with age,the size and number of uterine fibroids（P 0.05）;there was no correlation between PDGF expression and e IF4 E expression in surrounding normal uterine tissues（r = 0.324,P = 0.3950.05）,while PDGF expression was positively correlated with e IF4 E expression in uterine fibroids（r = 0.765,P = 0.016 0.05）.Conclusion The expressions of PDGF and e IF4 E in uterine fibroids are high,which are closely correlated with the related parameters of uterine fibroids,so PDGF and e IF4 E can be used to predict and diagnose the occurrence and development of uterine fibroids clinically.PDGF expression is positively correlated with e IF4 E expression in uterine fibroids,which provide new targets for developing drugs in treatment o
