摘　　要：目的明确白介素6(interleukin-6,IL-6)升高对肾小管上皮细胞脂质代谢因子脂肪分化相关蛋白(adipose differentiationrelated protein,ADRP)的影响。方法酶联免疫吸附实验(ELISA)检测糖尿病患者血清IL-6的表达,体外培养人肾小管上皮细胞(human renal proximal tubularcells,HKCs),给予IL-6刺激48h,免疫荧光和Western blot检测ADRP的表达。结果对糖尿病患者血清的检测显示:相对于健康体检者,糖尿病患者血清IL-6水平显著升高;IL-6刺激人肾小管上皮细胞48h后,免疫荧光检测发现ADRP主要表达在细胞质,荧光强度明显升高,Western blot结果也证实给予IL-6刺激后ADRP升高。结论 IL-6可上调肾小管上皮细胞的脂质标记因子ADRP,可能是引起糖尿病患者肾脏脂质代谢异常的因素之-。Objective To determine the effect of enhanced IL-6 on lipid metabolism factor ADRP in renal tubular cells.Methods ELISA was used to detect serum IL-6 in diabetic patients.The human renal tubular cells cultured in vitro were stimulated with IL-6 for 48 hours and then ADRP was detected by immunofluorescence and Western blot.Results Compared with normal control group,diabetic patients showed increased serum IL-6.The in vitro experiment revealed that ADRP was mainly located in cell plasma.The level of ADRP was significantly increased with the treatment of IL-6.Conclusion IL-6 can up-regulate ADRP in renal tubular cells,which may be one of the mechanisms involved in abnormal lipid metabolism of diabetic nephropathy.
