摘 要:利用广谱持久抗瘟基因Pigm的紧密连锁标记S29742,通过分子标记辅助选择和连续回交育种实践,定向改良早籼稻1701的稻瘟病抗性。获得以下主要结果:Pigm基因供体亲本谷梅4号与受体亲本1701对23份供试稻瘟菌菌株的抗性表现及抗菌谱差异显著,两亲本的抗性频率分别为95.7%和39.1%。S29742是一个基于PCR的共显性DNA标记,在谷梅4号和1701之间多态性明显且稳定,在两亲本中分别扩增出一条约550 bp和一条约900 bp的条带;S29742标记基因型对稻瘟病抗性表型的选择效率可达100%。通过连续回交、选择和自交,获得含Pigm基因的BC5F2群体,在此基础上通过基因型和表型分析鉴定出3个高抗稻瘟病的BC5F3株系,并通过田间主要农艺及产量性状分析,育成一个遗传背景与受体亲本相似的抗瘟水稻新品系"1701-Pigm"。To directionally improve blast resistance of the early indica rice line 1701, a molecular marker S29742 was screened, which is tightly-linked with the broad-spectrum blast resistance gene Pigm, and marker-assisted selection breeding practice was implemented in this study. The main results were as follows: the blast resistance performance and resistance spectrum to 23 M. oryzae isolates were quite different between the Pigm gene donor Gumei4 and receptor 1701, the resistance ratio were 95.7% and 39.1% respectively. S29742 is a co-dominant DNA marker based on PCR technique, showed obvious and stable polymorphism between Gumei 4 and 1701,produced one approximate 550 bp band and one 900 bp or so band from two parents respectively, and the selection efficiency of the S29742 marker genotype for blast resistance phenotype was 100%. The BC5F2 population harboring the Pigm gene was developed by means of consecutive backcrossing, selection and self-pollination, three BC5F3 family lines with high-level blast resistance were identified by genotypic and phenotypic analysis, and one blast resistant rice line possessing similar genetic background, named 1701-Pigm, was bred by field trial of main agronomic and yield traits.
