摘 要:为了明确糯质和非糯谷子Waxy基因c DNA序列变异,为品质育种提供实验依据,以2份糯质和2份非糯谷子为材料,利用特异RT-PCR和序列比对,研究了Waxy基因exon4~exon9区间c DNA序列SNPs及其氨基酸多态性,结果表明,糯质红粘谷(00000537)与白米粘谷的同源性为99.12%,在第163、第192、第245和第358位点检测到4个SNPs;非糯类型丰田501与赤谷4号的同源性为98.9%,在第139、第189、第282、第445和第451位点有5个SNPs。糯质红粘谷(00000537)与非糯品种赤谷4号相似性为98.23%,红粘谷(00000537)在第139、163、189、192、245、358、445和451位点碱基分别为T、G、T、G、G、A、C和A,赤谷4号在上述位点依次分别为C、A、G、A、A、C、T和G。糯质红粘谷(00000537)与非糯品种丰田501的相似性为98.9%,红粘谷(00000537)在163、192、245、282和358位点分别为G、G、G、A和A,丰田501依次分别为A、A、A、G和C。红粘谷(00000537)和赤谷4号exon4~exon9区间的c DNA序列编码的氨基酸序列中,检测出6个氨基酸变异,和丰田501出现3个氨基酸的变异。In order to make clear the c DNA sequence variations of waxy gene between waxy and no-waxy foxtail millets, and provide experimental basis of quality breeding, two waxy and two no-waxy foxtail millets were used to analyze the c DNA sequence single nucleotide and their amino acid polymorphisms by specific RT-PCR markers and sequence alignment. The results showed the homology between Hongniangu(00000537)and Baiminiangu was 99.12%, associated four single nucleotide polymorphism variations with 163 th, 192 th, 245 th and 358 th bases, respectively. Homology for Fengtian501 and Chigu4 was 98.9%, associated five single nucleotide polymorphisms with 139 th, 189 th, 282 th, 445 th and 451 th bases, respectively. The homology in Hongniangu(00000537)and Chigu4 was 98.23%, the single nucleotide polymorphism loci in the former were T, G, T, G, G, A, C and A, which were in 139 th, 163 th, 189 th, 192 th, 245 th, 358 th, 445 th and 451 th bases, respectively, but that in the latter were C, A, G, A, A, C, T and G in the same bases, respectively. The homology in Hongniangu(00000537) and Fengtian501 was 98.9%, the single nucleotide polymorphism loci in the former were G, G, G, A and A, which were in 163 th, 192 th, 245 th, 282 th and 358 th bases, respectively, but that in the latter were A, A, A, G and C in the same locis, respectively. Six amino acid variants were detected in amino acids sequence alignment, which were translated from c DNA sequence of Exon4~exon9 interval in Hongniangu(00000537) and Chigu4.
