摘 要:目的:研究脂氧素A4在睾丸缺血-再灌注损伤过程中对免疫细胞CD4^+ T的调节,保护生殖细胞功能作用。方法:健康成年大鼠40只随机分成A组(假手术组)、B组(睾丸扭转组)、C组(扭转复位组)、D组(扭转复位脂氧素A4治疗组),每组各10只。于术后24h分别检测患侧睾丸组织中IFN-γ、IL-2、IL-4、IL-10的浓度,以及睾丸组织中MDA、SOD、GPx含量。对各组睾丸组织行生殖细胞凋亡检测,行HE染色和Johnsen功能评分等形态学观察。结果:与A组相比,睾丸扭转后细胞因子IFN-γ、IL-2和组织中MDA显著升高(P<0.05),而SOD、GPx明显降低。与B和C组比较,D组细胞因子IL-4、IL-10,以及组织中SOD、GPx显著升高(P<0.05),细胞因子IFN-γ、IL-2和睾丸组织MDA水平降低明显。与B和C组比较,D组睾丸凋亡指数明显降低(P<0.05),Johnsen评分明显升高(P<0.05)。透视电镜下,B组和C组生殖细胞内线粒体内嵴结构破坏,溶酶体肿胀,细胞间隙增宽,D组细胞结构完整。结论:在睾丸缺血-再灌注过程中LXA4可以通过调节CD4^+ T细胞炎性因子的释放,减轻再灌注损伤炎症对生殖细胞的损伤,对生殖功能起保护作用。Objective:To study the immune regulation effect of Lipoxin A4(LXA4)on CD4+ T cells and the protective effect on germ cells in testicular ischemia and reperfusion injury in rats.Methods:Forty rats were randomly divided into 4groups:sham group(A);testicular torsion group(B);torsion/detorsion(T/D)group(C),T/D plus LXA4-pretreated group(D).Biochemical analysis(IFN-γ,IL-2,IL-4,IL-10,MDA,SOD and GPx in the testes),apoptosis assessment of germ cells,and morphologic evaluation were carried out after orchiectomies.Results:Compared with group A,the levels of proinflammatory cytokines(IFN-γ,IL-2)and MDA were significantly increased in the testes(P〈0.05),and the levels of SOD and GPx were decreased after testicular torsion.Compared with B and C,the levels of IL-4,IL-10 and tissue SOD/GPxwere significantly increased(P〈0.05)in group D,while IFN-γ,IL-2and MDA levels were significantly decreased.In addition,the AI(apoptosis index)values of group D was significantly lower than group B and C(P〈0.05),whereas Johnsen score was significantly increased(P〈0.05).Furthermore,swollen mitochondria with degenerated cristae and enlarged intercellular spaces were observed under transmission electron microscopy in group D.Conclusion:LXA4can regulate cytokine secretion of CD4~+ T cells and reduce the inflammation injure to germ cells,thus protecting reproductive function in testicular ischemia-reperfusion in rats.
