摘  要:目的观察冠通方对兔心肌缺血再灌注损伤后心肌细胞凋亡的病理变化,进一步证实冠通方对心肌细胞的保护作用。方法将48只新西兰大白兔随机分为假手术组、模型对照组、中药对照组、冠通方大剂量组、冠通方中剂量组、冠通方小剂量组,每组各8只。各组分别连续灌胃14 d,制备左室心肌病理切片。比较各组病理切片在光镜下心肌细胞病理形态学结构变化,使用原位末端标记法(TUNEL法)测定各组凋亡细胞指数。结果冠通方大、中剂量组心肌细胞在光镜下的结构形态得到较好的保护,冠通方大、中剂量组细胞凋亡指数较模型组显著下降(P<0.05)。结论冠通方可以减轻心肌缺血再灌注心肌细胞的损伤,有效减少心肌缺血再灌注后心肌细胞的凋亡,从而达到保护心肌细胞的作用。Objocllvo To observe the effect of Guantong decoction(GTD) on pathologic morpho logy in rabbits with myocardial ischmia reperfusion injury .Methods For ty -eight New Zealand white rabbits were randomly divided into 6 groups,namely sham grounormal control group,GTD high dose group, GTD moderate dose group,GTD low dose group and drug control group.The rabbits wetaken medicine for 14 days with designated drugs.GTG groups were taken different dose medicine of GTD,and sham operation grouand normal control group were pretreated by distilled water.The model of myocardial ischemia reperfusion injury were established bligating the left anterior descending artery of New Zealand rabbits for forty minutes after taken medicine for one hour and no coronaartery injury were in sham group.The heart were made into pathological sections and stained with HE and TUNEL.Apoptotic index wadetected with electron microscope in times of four hundreds.Results Myocardial injury cells and apoptotic index of GTD high dosgroup and moderate dose group were less than normal control group and GTD small dose group ( P 〈0.05) .Conclusion GTD caeffectively protect myocardial cell in rabbits with myocardial ischemia reperfusion injury.
