摘 要:基底拓扑结构是调控细胞生物学行为的重要因素之一。越来越多的研究表明人工构建的基底拓扑结构对工程化控制体外细胞培养的微环境、探究细胞与基底相互作用机理及医学植入物表面处理具有重要意义,然而,尽管关于该主题已发表了大量论文,但是由于细胞类型、基底材料、几何形状和测量参数等的差异,很难建立基底拓扑结构对细胞行为影响的显著趋势。本文梳理了基底拓扑结构的常用制造工艺,分析了不同基底拓扑结构对不同细胞的形态、迁移、增殖和分化的影响,讨论了可能存在的分子响应机理,以期对细胞力学的学术研究提供新的视角和基础资料。Substrate topography is one of the important factors that regulate the biological behavior of cells. More and more studies have shown that the artificially constructed substrate topography is of great significance for engineering control of the microenvironment of cell cultured in vitro, exploring cell-substrate interaction and related mechanics signal transduction pathways and medical implant surface treatment. However+ despite the large number of papers published on this topic, clear trends about the effects of substrate topography on cellular biological responses are difficult to establish, owing to differences n cell type, substrate material, geometrical shapes and measurement parameters. In this paper, the commonly used fabrication processes of substrate topographies were summarized, the effect of substrate topography on cell morphology, migration proliferation and differentiation were analyzed, and the possible mechanisms of response were discussed, with a view to providing new perspectives and basic data for the academic research of cell mechanics.
