摘　　要：目的观察Toll样受体4(TLR4)/核转录因子kappa B(NF-κB)信号通路及其相关炎症因子对周围性面神经炎的影响。方法 50例周围性面神经炎患者为试验组,50例同期健康体检者为对照组。用实时荧光定量聚合酶链式反应(RT-q PCR)和蛋白质印迹法(Western blot)检测外周血单核细胞(PBMCs)中TLR4 mRNA及蛋白;Western blot检测PBMCs中NF-κB p65磷酸;酶联免疫吸附法(ELISA)检测血清中肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)。结果试验组和对照组PBMCs中TLR4 mRNA的相对表达水平分别为1.55±0.25,0.83±0.10;TLR4蛋白的相对表达水平分别为1.11±0.17,0.61±0.11;试验组和对照组PBMCs中NF-κB p65磷酸化水平分别为0.67±0.30,0.36±0.22;TNF-α分别为(149.50±12.12),(102.10±10.60)ng·L-1;IL-1β分别(255.50±19.19),(105.20±12.80)ng·L^(-1),差异均有统计学意义(P<0.05)。结论 PBMCs中TLR4/NF-κB信号通路的活化及相关炎症因子的表达变化可能在周围性面神经炎的发生发展中发挥作用。Objective To explore the effect of toll- like receptor 4 （ TLR 4） / nuclear factor - kappa B （ NF - κB） pathway and its related inflammatory markers on peripheral facial neuritis. Methods Fifty patients with peripheral facial neuritis were enrolled in the treatment group, and fifty healthy subjects were enrolled in the control group. Real -time fluorescent quantitative PCR （RT -qPCR） and Western blot was used to detect the expression of TLR4 mRNA and protein. Western blot were used to detect NF - κB p65 phosphorylation in peripheral blood mononuclear cell （PBMCs）. Enzyme linked immunosorbent assay （ELISA） was used to detect the content of serum tumor necrosis factor - α（ TNF - α） and interleukin - 1β （ IL - 1β）. Results The relative expression levels of TLR4 mRNA in PBMCs of treatment group and control group were 1.55 ± 0.25 and 0.83 ±0. 10, while that of TLR4 protein were 1.11 ±0. 17,0. 61 ±0. 11 ; the phosphorylation levels of NF - κB p65 in treatment group and control group were 0. 67 ：t： 0. 30, 0. 36 ± 0. 22 ; the serum TNF-α in the treatment group and the control group were （149.50±12.12）, （102.10±10.60） ng L-1, while the serumIL - 1β were （255.50 ± 19. 19）, （ 105.20 ± 12.80）ng · L^-1 ,the differences were statistically significant （P 〈0. 05）. Conclusion The expression changes of TLR4/NF - κB pathway in peripheral blood mononuclear ceils and related inflammatory markers in serum may play a role in peripheral facial neuritis.
