摘 要:目的研究色满酮类衍生物(HEF-19)对小鼠的止泻作用。方法采用蓖麻油、硫酸钠、液体石蜡诱导的腹泻模型,通过测定稀便率、稀便级和腹泻指数研究HEF-19(300、100、50 mg·kg^(-1))的止泻作用,通过对正常小鼠的胃排空、小肠推进实验和离体平滑肌实验探讨其可能的作用机制。结果HEF-19各剂量组灌胃给药与模型组相比可明显降低3种腹泻模型小鼠的稀便率、稀便级和腹泻指数;可明显抑制正常小鼠的小肠推进率、胃排空率。HEF-19(1×10^(-5)~1×10^(-3)mol·L^(-1))可显著抑制CaCl_2诱导的离体小肠平滑肌收缩。结论 HEF-19可能是通过阻断钙离子通道,抑制胃肠运动起到止泻作用。Objective To determine the antidiarrhea effect of a chromane derivative(HEF-19) on mice. Methods The diarrhea model induced by castor oil, sodium sulfate and liquid paraffin was used. The loose stool rate, the diarrhea degree and the diarrhea index were studied to determine the antidiarrhea effect of HEF-19(300, 100 and 50 mg·kg^-1), and to explore the possible mechanism of HEF-19 through gastric emptying, small intestine propulsion and isolated smooth muscle experiment. Results Compared with the model group, each dose of HEF-19 significantly reduced the rate of loose stool, the degree of diarrhea and the diarrhea index in mice. The small intestinal propulsion rate and gastric emptying rate were significantly inhibited by HEF-19 in normal mice. HEF-19(1×10^-5-1×10^-3mol·L^-1) significantly inhibited the contraction of isolated small intestinal smooth muscle induced by CaCl2. Conclusion The antidiarrheal effect of HEF-19 is through blocking calcium channels and inhibiting the gastrointestinal motility.
