摘 要:目的测定坚杆火绒草及其不同炮制品中绿原酸、咖啡酸和木犀草素的含量。方法采用Acquity UPLC BEH C18色谱柱(50 mm×2.1 mm,1.7μm),流动相A为甲醇、B为0.1%磷酸水溶液,梯度洗脱,柱温35℃,流速0.2 m L·min^(-1),进样量1μL,多波长分段检测。结果绿原酸、咖啡酸和木犀草素的线性范围分别为:3.68~73.60 ng(r=0.9999)、1.00~20.00 ng(r=0.9999)、0.90~40.50 ng(r=0.9999)。坚杆火绒草中绿原酸、咖啡酸和木犀草素的含量差异较大,同一植株的叶、花和果实中绿原酸和咖啡酸的含量明显高于根和茎中的;根和茎中均未检测到木犀草素,而花和果实中均含有木犀草素。结论藏医火灸柱水煮后晒干的工艺不建议继续使用。OBJECTIVE To determine the content of chlorogenic acid, caffeic acid and luteolin in Leontopodium franchetii Beauv. and in its different processed products. METHODS An Acquity UPLC BEH C iscolumn (50 mm× 2.1 mm, 1.7 μm) was used at the temperature of 35℃. The mobile phase consisted of methanol and 0.1% phosphoric acid of gradient elution with the flow rate of 0. 2 mL. min- l , and the injection volume was 1 puL, and the detection wavelength was segmented. RESULTS The linear ranges of chlorogenic acid, caffeic acid and luteolin were 3.68 - 73.60 ng ( r = 0. 9999 ) , 1.00 - 20.00 ng ( r = 0. 9999 ) and 0.90 - 40.50 μg (r = 0. 9999 ), respectively. There were significant differences in the content of chlorogenic acid, caffeic acid and luteolin in L. franchetii. The contents of chlorogenic acid and caffeic acid in the flower and fruit parts were higher than that in the root and stem parts. The luteolin was not detected in the root and stem parts, but were detected in flower and fruit parts. CONCLUSION It suggests that the boiling - dry process of Tibetan fire moxibustion should be abolished.
