摘 要:目的研究脾气虚大鼠焦虑程度的变化并探讨其可能机制。方法选取雄性SD大鼠16只,体质量(200±10)g,将其随机分为对照组和脾气虚组(模型组),每组8只。旷场实验评价大鼠焦虑程度;ELISA法测定海马组织ATP和缩胆囊素-8(CCK-8)含量;免疫组化法检测海马CCKB型受体(CCKBR)表达。结果与对照组比较,模型组的焦虑程度显著降低(P<0.01),海马组织的ATP含量降低(P<0.05),海马CCK-8水平变化不明显(P>0.05),但其受体表达下降(P<0.05)。结论脾气虚时焦虑程度下降,其初步机制与海马能量不足和CCKBR表达减少有关。Objective To study the anxiety degree change of rats in spleen qi deficiency rat and discuss its initial mechanism.Methods 16 male SD rats with the body mass of (200±10)g were selected and they were randomly divided into the control group and spleen qi deficiency group (model group),and there were 8 rats in each group.Open-field test (OF1,) was used to evaluate anxiety degree of rats.ELISA was chosen to measure ATP and CCK-8 concentrations in hip pocampal tissue.Immunohistrochemical method was used to check CCKBR expression in hippocampal tissue.Results Compared with those in the control group,rat's anxiety was reduced (P〈0.01),hippocampal ATP level was decreased (P〈 0.05),hippocampal CCK-8 concentration was unchanged (P〉0.05),hippocampal CCKBR expression was decreased (P〈 0.05) in the model group.Conclusion Anxiety in spleen qi deficiency is lower and its initial mechanism might involve in decrease of hippocampal ATP level and CCKR expression.
