摘  要:目的优化前脂肪细胞的分离培养方法 ,观察其形态学变化,比较其增殖以及成脂分化的潜能。方法无菌条件下采集腹部开放手术患者的网膜脂肪及腹腔内肾周脂肪,经胶原酶消化法分离培养人前脂肪细胞,倒置显微镜下观察人前脂肪细胞的形态,对传代至第3代的前脂肪细胞通过绘制生长曲线比较增殖能力的差别,并进行成脂诱导分化后,采用油红O染色的方法对其分化能力进行鉴定。结果不同部位获得的前脂肪细胞形态以长梭形为主,增殖活跃,其中腹腔内前脂肪细胞数量更多,呈旋涡状生长。腹腔内前脂肪细胞的增殖能力高于网膜来源的细胞。经成脂诱导分化后,可以诱导为成熟的脂肪细胞。结论采用腹腔内脂肪组织得到的人前脂肪细胞在体外易于分离培养,数量较多、纯度较高、活性更好,可以稳定传代。此种方式获得的前脂肪细胞可以为研究肥胖等相关代谢性疾病提供实验室基础。Objective To optimize the isolation and culture of preadipocytes, and to determine the morphologi- cal changes, and to compare their proliferation and the potential of adipogenic differentiation. Methods The omental fat and intraperitoneal perirenal fat were obtained from patients undergoing open surgery under sterile condition. The human preadipocytes were isolated and cultured by collagenase digestion. The morphology of human preadanocytes was examined under inverted microscope. By drawing the growth curve, the difference in the proliferation of the third passage preadipocytes was compared. The adipogenic differentiation of the third passage preadipocytes was performed, and the differentiation was identified by oil red O staining. Results The preadipocytes obtained from different parts were mainly spindle-shaped, and the proliferation was active. The number of the intraperitoneal preadipocytes was large, with a spiral-like growth. The proliferation of preadipocytes in the abdominal cavity is higher than that of the o- mentum cells. After adipogenic differentiation, the preadipocytes can be induced into mature adipocytes. Conclusion The experimental data show that the human preadipocytes obtained from the intraperitoneal adipose tissue are easy to be isolated and cultured in vitro, with large number, high purity, good activity, and stable passage. The preadipocytes obtained in this way can provide a laboratory basis for the study of metabolic diseases, such as obesity.
