摘 要:以绿玉菊的叶片和带腋芽茎段为试材,以MS为基本培养基,研究不同激素配比培养基对组织快繁的影响以及外植体最适消毒组合。结果表明,叶片的最适诱导培养基为MS+NAA 0.2 mg/L+6-BA 3.0 mg/L,出愈率达80%;带腋芽茎段的最适诱导培养基为MS+NAA0.1 mg/L+6-BA 1.0 mg/L,诱导率高达92%;最适生根培养基为MS+NAA 0.05 mg/L,生根效果最佳、根系粗壮、次级根多;外植体消毒宜采用酒精和升汞(20 s+7 min)的搭配,污染率可以控制在15%。To study the effects of different hormone ratios media on the rapid propagation of tissue and the optimal disinfection combination of explants with leaves and stem segments with axillary bud of Senecio macroglossus and MS as the basic medium.The results showed that the optimum medium was MS+NAA 0.2 mg/L+6-BA 3.0 mg/L,and the callus rate was 80%.The optimum medium for induction of axiUary buds was MS+NAA 0.1 mg/L+6-BA 1.0 mg/L,the induction rate was up to 92%.The optimum rooting medium was MS+NAA 0.05 mg/L,the root was thick and more secondary root.The combination of alcohol and mercury(20 s+7 min)was the best suited for disinfecting explants,which could control pollution rate to 15%.
