摘 要:为研究不同方法测定大豆花粉活力的问题,选取‘绥农4’、‘吉林47’和‘CK-P’为研究对象,通过蔗糖、琼脂、硼酸、温度单因子正交试验筛选出的培养基进行离体萌发,比较I2-KI染色法、TTC染色法和离体萌发法3种花粉活力测定方法的不同。结果表明:I2-KI染色法容易操作,因这种方法较离体萌发法简单,测定结果比较准确,成本低,所需时间短,常用来观测大豆花粉活力。TTC法染色效果与I2-KI染色法接近,没有差异,也可作为检测大豆花粉活力的方法。离体萌发法测定大豆花粉活力的方法和很多因素有关,与I2-KI染色法和TTC染色法有显著差异,且其培养基的配制繁琐,所以用I2-KI染色法和TTC染色法来测定大豆的花粉活力。To study different methods for determining soybean pollen vitality, 'Suinong4', 'Jilin47' and 'CK- P' were used as the materials, a single factor orthogonal test was conducted based on sucrose, agar, H3BO3 and temperature, to screen culture medium for pollen germination, and the difference of pollen viability under I2- KI, TTC staining and in vitro germination were compared. The results showed that the I2-KI staining method was easy to operate, and was simpler than the in vitro germination method, its determination result was accurate, the cost was low and the time was short, so it was suitable for testing pollen vitality. TTC staining was similar to 12-KI staining, and could also be used as a method to detect soybean pollen vitality. The method of in vitro germination of pollen viability was related to many factors, and was significantly different from I2-KI and TTC staining, and its preparation was complex, so I2-KI and TTC staining method could be used to determine soybean pollen viability.
