摘 要:目的:观察虎纹蜘蛛毒素(HWTX-I)对运动性骨关节损伤大鼠软骨诱导型一氧化氮合成酶(iNOS)的表达及血清中一氧化氮(NO)含量的影响,探讨HWTX-I对慢性运动性骨关节损伤的保护作用机制,为其临床应用提供理论和实验依据。方法:雄性SD大鼠30只,采用改良Hulth模型结合间歇性中低强度运动建立大鼠运动磨损性骨关节病理模型。随机分成3组:非用药组、HWTX-I用药组和布洛芬用药组。对各组大鼠膝关节软骨进行显微结构形态学观察,采用免疫组织化学和图像分析系统检测iNOS水平的差异表达,分光光度法测定血清中NO含量。结果:3组iNOS的表达强度相比较,HWTX-I用药组明显低于非用药组和布洛芬用药组;HWTX-I用药组血清中NO的含量亦明显低于非用药组和布洛芬用药组。结论:INOS、NO在运动性骨关节损伤的病理进程中具有重要作用,HWTX-I可通过下调其在关节软骨的表达和血清中的含量,对大鼠运动性骨关节损伤关节有一定的抗损伤保护作用。Objective The effects of Huwentoxin-I(HWTX-I) on iNOS experssion in articular cartilage of the rat chronic sports arthritic injury and the changes of NO concentration in serum,the mechanism of HWTX-I on protecting sports arthritis against injury is researched,and had provided theoretical and experimental basis for its clinical application.Methods Thirty male SD rats were randomly divided to three groups,which is Nonadministered group,HWTX-I administered group and Ibuprofen administered group.By adopting the improved Hulth model combined with intermittent low-intensity exercise,all SD rats were established the rat sports durable osteoarthropathy pathological model.Observing the morphological change of the Knee cartilage,all samples were undertaken immunohistochemistry and image analysis methods in order to test their iNOS expressing intensity.Spectrophotometry was used to detect the concentration of NO in serum.Results The expressing of iNOS in HWTX-I administered group was significantly different from Non-administered group and the Ibuprofen administered group,moreover,the concentration changes of NO in serum of HWTX-I administered group was significantly different from Non-administered group and Ibuprofen administered group.Conclusion INOS and NO takes a important role in the pathological process of sports arthritic injury.HWTX-I has apparently antinociceptive efficacy on sports arthritic injury by down-regulating the expression of iNOS and the concentration of NO in serum.
