摘  要:为检测禽波氏杆菌鸡胚分离株和呼吸道分离株对雏鸡的致病性差异,本研究对2004年~2011年分离鉴定的12株禽波氏杆菌进行了鸡胚半数致死量和雏鸡半数致死量的检测,筛选强毒菌株进行雏鸡的鼻腔感染试验。结果表明LL、XT和AQ分离株的毒力最强,其它分离株的毒力稍弱。3株强毒菌株均可感染雏鸡呼吸道和肺脏,引起呼吸道症状以XT和AQ的症状最为明显,而在其它内脏中造成感染的鸡只比例以LL株最高,可达30%~40%;AQ最低,仅10%。HE染色结果显示对气管的损伤以XT最强,而肺脏、肝脏等内脏器官的病理变化以LL最严重。免疫酶组化试验也证实LL在内脏组织中的载量要多于XT和AQ。3株菌株均可导致雏鸡的生长抑制和免疫器官损伤,对新城疫灭活苗免疫后的抗体水平具有轻微的抑制作用,且均以LL的抑制作用最强。实验结果显示在所检测的12株禽波氏杆菌中,鸡胚分离株LL毒力最强,具有比呼吸道分离株XT和AQ更强的感染雏鸡内脏组织的能力,本研究为进一步鉴定禽波氏杆菌毒力因子和免疫原性奠定了基础。Bordetella avium can cause a highly contagious disease of upper respiratory tract of poultry. It's usually isolated from chick respiratory tracts or chick embryos. However, the differences between them are still uncertain. Twelve Bordetella avium strains were isolated and identified from 2004 to 2011 in this study. Virulent isolates were screened to challenge chicks by intranasal based on results of ELDs0 and LDso. It was found that all the 3 isolates could infected respiratory tracts and lungs of chicks. XT and AQ caused the most serious respiratory symptoms, however, LL caused the highest infection rate, up to 30%-40%, in visceral. AQ caused the lowest, only 10%. The results were fiu'ther illustrated by HE staining and immunoenzymatic histochemistry detection. XT caused tracheal hurt. LL caused the most serious pathological changes in the lung, liver and other visceral. A large number of bacteria were detected from organ tissues, too. Furthermore, LL had the most effect on the growth of chick, immune organs, and antibody titers of New castle disease after immunization by inactivated vaccine. In conclusion, LL, as a chick embryo isolate, has the strongest virulence among 12 Bordetella av/tun isolates tested in this study. It has more stronger damage in visceral organs than isolate XT and AQ from chicks. The results laid a foundation for further identification of its virulence factors and immunogenicity.
