植酸对结直肠癌模型大鼠zo-1、occludin和claudin-1蛋白表达的影响  被引量:7

摘  要:目的观察不同剂量植酸干预对结直肠癌模型大鼠zo-1、occludin和claudin-1蛋白表达的影响,探讨植酸对结直肠癌大鼠肠黏膜屏障的作用。方法将60只Wistar雄性大鼠按体质量随机区组分为正常对照组(NC组)、模型组(MG组)、植酸低剂量组(PL组)、植酸中剂量组(PM组)和植酸高剂量组(PH组)。PL、PM和PH组大鼠每天分别给予0.25、0.50和1.00g/kg的植酸钠灌胃,NC和MG组大鼠以等体积的生理盐水代替。从实验第5周起,除NC组外,其余各组大鼠于颈背部注射剂量为30mg/kg的二甲肼,持续18周,期间NC组大鼠于颈背部注射等体积的生理盐水。继续植酸钠灌胃至实验第39周末,处死大鼠。取NC组大鼠结肠组织及其他组大鼠结直肠肿瘤组织,采用免疫组化方法检测zo-1和occludin蛋白的表达,采用Western Blot方法检测claudin-1蛋白的表达。结果 MG组大鼠结直肠肿瘤发生率为100%,PL、PM、PH组大鼠结直肠肿瘤发生率分别为100%、88.9%和66.7%。与NC组比较,MG组大鼠zo-1和occludin蛋白的表达明显降低(F=23.118、52.886,t=9.041、13.050,P<0.05);与MG组比较,PL、PM、PH组大鼠zo-1和occludin蛋白的表达显著升高(t=2.385~8.611,P<0.05)。与NC组比较,MG组大鼠claudin-1蛋白的表达明显升高(F=67.159,t=14.028,P<0.05);与MG组比较,PM和PH组大鼠claudin-1蛋白的表达显著降低(t=2.635、8.421,P<0.05)。结论植酸可以通过调节zo-1、occludin和claudin-1蛋白的表达来改善肠黏膜屏障,从而抑制结直肠癌的发生。Objective To observe the effects of different doses of phytic acid (PA) on the expression of zo-1, occludin, and claudin-1 in rats with colorectal cancer (CRC), and to investigate the effects of PA on the intestinal mucosal barrier in rats with CRC. Methods Sixty male Wistar rats were equally and randomly divided into five groups: normal control group (NC group), model group (MG group), low-close PA group (PL group), middle-dose PA group (PM group), and high-dose PA group (PH group) according to their body weight. The PL group, PM group, and PH group were given sodium phytate at 0.25, 0.50 and 1.00 g/(kg·d), respectively, by gavage, while the NC group and MG group were given an equal volume of normal saline. From the 5th week to the 23rd week, all rats except those in the NC group were injected subcutaneously with dimethylhydrazine (30 mg/ kg) in the back of the neck to induce CRC; the NC group received an equal volume of normal saline. All rats were sacrificed at the end of the 39th week. The colon tissue and the CRC tissue were collected from the rats in the NC group and the other groups, re spectively, to measure the expression of zo 1 and occludin by immunohistochemistry and the expression of claudin-1 by Western blot. Results The incidence rates of CRC in MG group, PL group, PM group, and PH group were 100%, 100%,88.9%, and 66.7 %, respectively. Compared with the NC group, the MG group had significantly lower expression of zo-1 and oceludin (F = 23.118,t 9.041;F=52.886,t=13.050;P〈0.05). Compared with the MG group, the PL group, PM group, and PH group had significantly higher expression of zo 1 and oecludin (t =2.385--8.611 ,P〈0.05).
