摘 要:目的通过BN大鼠皮肤贴敷和皮内注射试验染毒具有致敏性的外源性化学物质己基肉桂醛,观察致敏特点,同时以豚鼠作为对照观察动物,探讨BN大鼠作为化学品过敏试验动物模型的可能性。方法将BN大鼠和白色豚鼠分别随机分为2组:对照组和己基肉桂醛组,每组10只。将己基肉桂醛组实验动物于试验的第1天分别进行皮肤贴敷试验和皮内注射试验,皮肤贴敷试验于首次贴敷后28 d进行激发试验,皮内注射试验于皮内注射后21 d进行激发试验;对照组给予相同体积的生理盐水注射液。激发后观察每只动物过敏反应情况,根据过敏反应症状评分,计算致敏率,对致敏强度进行分级。经腹主动脉取血,分离血清,竞争ELISA法检测血清Ig E含量。结果皮肤贴敷试验中染毒己基肉桂醛后,BN大鼠和豚鼠致敏率分别为60%和50%;BN大鼠血清Ig E含量与对照组相比差异有统计学意义(P<0.05),豚鼠Ig E含量与对照组相比差异无统计学意义(P>0.05)。皮内注射试验中染毒己基肉桂醛后,BN大鼠和豚鼠致敏率分别为70%和80%;BN大鼠和豚鼠己基肉桂醛组血清Ig E含量与各自的对照组相比,差异有统计学意义(P<0.05)。结论在皮肤贴敷和皮内注射过敏反应中,BN大鼠已基肉桂醛组呈阳性反应,染毒后的局部反应特点强于豚鼠,因此,BN大鼠作为化学品过敏试验的动物模型具备可能性。Objective To infect BN rats with xenobiotics hexylcinnamaldehyde, an allergic exogenous chemical compound, by intradermal injections and skin sticking, and observe the characters of allergic reaction. Meanwhile, the possibility of of BN rats as an animal model of allergy test for chemicals was investigated. Methods Both BN rats and white guinea pigs were randomly divided into control and test groups, 10 for each. Skin sticking test and intradermal injection test were performed in test group on day 1, while challenge test was performed 28 d after the first skin sticking test and 21 d after intradermal injection test. The animals in control group were treated with the same volume of physiological saline. After the challenge test, the allergic reaction of each animal was observed and scored, based on which the sensitization rate was calculated, and the reaction was graded. The blood samples were collected from the aorta abdominals, and the sera were separated and determined for Ig E content by competitive ELISA. Results The sensitization rates in BN rats and guinea pigs by skin sticking were 60% and 50% respectively. The serum Ig E content of rats in test group after infection by skin sticking showed significant difference with that in control group(P 〈 0. 05), while that in guinea pigs showed no significant difference(P 〉 0. 05). The sensitization rates of BN rats and guinea pigs by intradermal injection were 70% and 80% respectively, while both the serum IgE contents in BN rats and guinea pigs showed significant difference with those in control groups(each P 〈 0. 05). Conclusion BN rats were more sensitive to xenobiotics hexylcinnamaldehyde than guinea pigs in both skin sticking and intradermal injection tests, indicating the possibility of the rats as an animal model of allergy test for chemicals.
