抗癌扶正方不同提取液对顺铂化疗荷Lewis肺癌小鼠干预作用的实验研究

摘　　要：目的:观察抗癌扶正方水提液与醇提液对顺铂化疗荷Lewis肺癌小鼠的干预作用。方法:将制得的Lewis肺癌细胞接种至C57BL/6小鼠右侧腋窝皮下复制模型。实验随机分为5组:正常对照组、模型组、顺铂组、水提+顺铂组、醇提+顺铂组,每组10只,连续给药24 d。给药期间观察各组小鼠的一般反应,每4 d称量一次体质量和肿瘤体积。处死小鼠取血及脾脏,计算脾脏指数,检测白细胞计数;测量肿瘤体积和瘤重,观察肺组织病理变化情况。结果:药物干预24 d后,水提+顺铂组小鼠精神萎靡、反应迟钝等现象明显改善;肿瘤体积、瘤重及脾脏指数明显减小(P<0.05,P<0.01);体质量、白细胞数目明显升高(P<0.05,P<0.01);给药各组小鼠肺组织病理检查均显示瘤细胞出现灶状坏死区及核碎裂象,并有炎性细胞浸润,瘤组织内及其周围微血管减少。醇提+顺铂组小鼠上述各项指标均无明显变化。结论:抗癌扶正方水提液可明显改善抗癌药顺铂对荷Lewis肺癌小鼠的副作用,同顺铂配合使用具有协同抗癌作用;抗癌扶正方醇提液则无此作用。Objective： To observe the intervention effects of water and alcohol extract liquid of Kangai Fuzheng Fang on Lewis lung carcinoma mice treated with cisplatin. Methods： The obtained inoculated Lewis lung carcinoma cells were inoculated to C57BL/6 mice （half male and half female） right armpits. Randomly di- vided into 5 groups： normal control group, model group, cisplatin group, water extraction combined with cisplatin group, alcohol extraction combined with cisplatin group with each 10 mice, mice in each group were administered continuously for 24 days. The general reaction was observed in the nude mice of each group during the administration period, body weight and cancer weight were weighed every four days, cancer and spleen weight were weighed after sacrificed , the spleen index was calculated, leukocyte count was determined, pathological changes of lunge tissue were observed by light microscope. Results： After 24 days intervention of water and alcohol extract liquid of Kangai Fuzheng Fang, Compared with the model group and cisplatin group, water extraction combined with cisplatin group mice apathetic, unresponsive and so significantly improved; tumor volume, tumor weight and spleen index were significantly reduced （ P 〈 O. 05 ） ; body weight and white blood ceils increased significantly （ P 〈 0. 05 ） ; mice＇ s lung tissue pathological changes showed tumor cells appear spotty necrosis and nuclear fragmentation, inflammatory cell infiltration and microvascular surrounding tissue reduced in tumor, alco-hol extraction combined with cisplatin group showed no significant changes. Conclusion： The water extraction of Kangai Fuzheng Fang can significantly improve the side effects of anti - cancer drug cisplatin on Lewis lung canc- er mice. Used in conjunction with cisplatin have synergistic anticancer effects., while the alcohol extraction of Kangai Fuzheng Fang has no effect of the side effects.
