摘 要:目的建立苗药夜关门药材的定性、定量检测方法,并制定质量标准。方法对苗药夜关门药材进行性状鉴别、显微鉴别,采用薄层色谱(TLC)法对其中的槲皮素进行定性鉴别,采用高效液相色谱(HPLC)法对其中的槲皮素进行含量测定;参照《中华人民共和国药典》2015版一部附录方法对苗药夜关门药材的水分、总灰分、醇溶性浸出物含量进行测定。结果薄层色谱斑点清晰,专属性强,且阴性无干扰;含量测定中,槲皮素的回归方程为Y=1 114.5X+43.507 2,r=0.999 9,结果表明槲皮素在0.128~1.280μg范围内线性关系良好;平均回收率为100.37%;初步预定苗药夜关门药材水分不得超过10.0%,总灰分不得超过6.0%,醇溶性浸出物含量不得少于12.0%。结论本法操作简单、结果可靠,可有效控制苗药夜关门药材的质量。Objective To establish qualitative and quantitative test methods for Lespedeza cuncata (Dum. Cours. ) G. Don. (Miao national herb) and work out the quality standard. Methods Morphological identification and microscopic identification of Lespedeza cuncata (Dum. Cours. ) G. Don. were made. Quercetin in it was quali- tatively identified by TLC. Content of quercitrin was determined by HPLC. The water content, total ashes, ethanol- soluble extractives were determined according to the approaches listed in Chinese Pharrnaeopeoa published in 2015. Results The TLC spots were clear without interference and had a strong specificity. The content of quercetin was in good line with the range of 0. 128 to 1. 280 p.g, Y= 1 114.5X+43. 507 2, r=0. 999 9. The average recovery was 100.37%. It was initially proposed that the content of moisture was at most 10.0% , total ash was no more than 6.0%, ethanol-soluble extractives was at most 12.0%. Conclusion The method is simple, reliable and can effec- tively control the quality of medicinal materials.
