摘 要:目的探讨丙泊酚麻醉对大鼠学习记忆能力及海马γ-氨基丁酸转运体-1(γ-aminobutyric acid transporter-1,GAT-1)表达的影响。方法健康雄性SD大鼠30只,4月龄,体重250~300g,采用随机数字表法将其分为3组(n=10):对照组(C组)、低剂量组(L组)和高剂量组(H组)。L,H组大鼠经尾静脉持续输注丙泊酚2h,于麻醉后24h进行Morris水迷宫测试,测试结束后立即灌注取脑,采用免疫组织化学法检测大鼠海马CA1区GAT-1的表达。结果与C组比较,L,H组大鼠逃避潜伏期延长,穿越原平台次数减少,海马CA1区GAT-1阳性细胞数增加(P<0.05);与L组比较,H组大鼠逃避潜伏期延长,穿越原平台次数减少,海马CA1区GAT-1阳性细胞数增加(P<0.05)。结论丙泊酚麻醉可导致大鼠学习记忆能力减退,其机制可能与海马GAT-1表达上调有关。Objective To investigate the effects of propofol on cognitive function and expression of γ-aminobutyric acid transporter-1(GAT-1) in hippocampus of rats.Methods Thirty healthy male SD rats,aged 4 months,weighing 250 ~ 300 g,were randomly divided into 3 groups(n=10 each) used a random number table:control group(group C),low dose group(group L) and high dose group(group H).The rats were anesthetized with propofol intravenously through the tail vein for 2 hours in group L and H.At 24 hours after anesthesia,Morris water maze test was used to assess the cognitive function,and the rats were killed immediately after the test.The expression of GAT-1 in the hippocampus CA1 region was detected by immunohistochemistry.Results Compared with group C,the escape latency was significantly prolonged,the frequency of crossing the original platform was decreased,and the number of GAT-1 positive cells in the hippocampal CA1 region were increased after anesthesia in L and H groups(P〈0.05);compared with group L,the escape latency was significantly prolonged,the frequency of crossing the original platform was decreased,and the number of GAT-1 positive cells in the hippocampal CA1 region were increased after anesthesia in group H(P〈0.05).Conclusion Propofol anesthesia may result in cognitive dysfunction in rats,and its mechanism is likely to be related with up-regulation of GAT-1 expression in hippocampus.
