摘 要:目的:研究黄管秦艽中龙胆苦苷对骨关节炎大鼠的治疗作用。方法:Wistar大鼠关节腔内注射造模剂造模,抽检确定造模成功后,将造模大鼠分为模型组、双醋瑞因(9 mg/kg)阳性对照组及龙胆苦苷低、中、高(50、100、200 mg/kg)剂量组,另设正常对照组,每组10只。各组大鼠灌胃给药,10 m L/kg。4 w后处死动物,取胸腺、脾脏,计算脏器指数;取血测全血粘度;制备血清测定IL-1β、PGE2、COX-2、COX-1、NO含量;摘取膝关节,制作病理切片HE染色观察组织病理学改变。结果:与正常对照组比较,模型组大鼠胸腺指数显著升高(P<0.05),全血低、中、高切粘度显著升高(P<0.01),血清IL-1β、PGE2、NO、COX-2含量显著升高(P<0.05或P<0.01);与模型组比较,各给药组胸腺指数显著降低(P<0.05),全血粘度显著降低(P<0.01),血清中IL-1β、PGE2、NO、COX-2含量显著降低(P<0.05或P<0.01);病理切片显示,与正常对照组比较,模型组关节软骨细胞排列紊乱且明显增生,染色严重降低,各给药组关节软骨细胞排列紊乱,染色中度降低。结论:龙胆苦苷可通过降低全血粘度及血清中IL-1β、PGE2、NO、COX-2含量起到对大鼠骨关节炎的治疗作用。Objective:To study the therapeutic action of gentiopicroside in Gentiana officinalis on the model rat.Methods:The osteoarthritis rat model was built by intraarticular injection papain plus L-cysteine.50 rats were randomly into five groups as model group,positive control group (diacerein 9 mg/kg),the low dose (50 mg/kg) group,the middle dose (100 mg/kg) group and the high dose (200mg/kg) group,with ten rats in each group,and there were also ten healthy rats as normal control group.Rats in each group were administered 10 mL/kg corresponding drugs in lavage.After four weeks of treatment,animals were sacrificed,the thymus gland and spleen were collected for organ index calculation.The blood were collected for the blood viscosity detection.The levels of IL-1β,PGE2,COX-2,COX-1,NO in the serum were detected by ELISA method.The pathological changes of articular cartilage were observed by HE staining.Results:Compared with the normal control group,the thymus organ index,the blood viscosity and the content of IL-1β,PGE2,NO,COX-2 in the serum were all significantly increased in the model groups.Compared with the model group,the thymus organ index,the blood viscosity and the content of IL-1β,PGE2,NO,COX-2 in the serum were significantly decreased in all medication administration groups.Compared with the normal control group,the cells of articular cartilage derangement and hyperplasia significantly,and the staining severely reduced in the model group,the cells of articular cartilage derangement and the staining midrange reduced.Conclusion:The gentiopicroside in G entiana officinalis has therapeutic action on the osteoarthritis model rats by decreasing the blood viscosity and the content of IL-1β,PGE2,COX-2,NO in the serum.
