补肾调肝方对多囊卵巢模型大鼠血清TGF-β1、IGF-1、TNF-a及激素水平的影响

摘　　要：目的:探讨补肾调肝方对多囊卵巢模型大鼠血清TGF-β1、IGF-1、TNF-a及激素水平的影响。方法:选取雌性大鼠(SD)45只,随机分为PCO模型组36只和正常对照组9只。参照Poretsky法,将造模成功的36只模型随机分为4个组,各9只。分别给予补肾调肝汤68.66g/kg(中药大剂量组)、17.17g/kg(中药小剂量组),二甲双胍250 mg/kg(二甲双呱组)和蒸馏水1ml/100g(对照组)体重灌胃,1次/d,连续用14d。放射免疫法检测T、E_2、LH、FSH、INS水平;用ELISA法检测TGF-β1、IGF-1、TNF-a水平。结果:中药大剂量组与其他组比较,T水平明显降低,E_2水平、E_2/T比值明显升高,血清TGF-β1、IGF-1水平升高,TNF-a水平降低,比较差异均有统计学意义(P<0.05)。结论:补肾调肝方能降低PCO模型大鼠血清T水平,升高E_2水平,也能够调节垂体LH、FSH的分泌,同时能够使PCO模型大鼠的血清TNF-a水平降低,TGF-β1、IGF-1水平升高,在一定程度上对恢复PCO模型大鼠的生殖功能有促进作用。Objective： To investigate the effects of Bushen Tiaogan decoction on serum TGF-β1 , IGF-1, TNF-a and hormone levels in rats with polycystic ovary syndrome （PCOS）.Methods： Female rats （SD）, 45 were randomly divided into 36 PCO model group and control group 9.Reference poretsky method, the successful model 36 models were randomly divided into 4 groups, each 9 were given the decoction of Bushen Tiaogan and the liver 68.66 g/kg （high-dose group）, 17.17 g/kg （small groups of Chinese medicine）, 250 mg/kg metformin （metformin Gung）, 1 ml/100g and distilled water （control group） weight loss prednisone 1/d, 14 d in a row.RIA T, E2, LH, FSH, INS level;By ELISA method for detecting levels of TGF-beta 1, IGF-1, TNF-a.Results： The high-dose group compared to other groups, T levels were significantly lower E2, E2/T ratio increased significantly increased serum levels of TGF-beta-1, IGF-1, TNF-a levels, there was a significant difference （P〈0.05）.Conclusion： Bushen Tiaogan decoction can reduce the level of serum T in PCO model rats, and can increase the level of E2 ,can also regulate the secretion of pituitary LH, FSH, at the same time, the serum TNF-a level of PCO model rats is decreased,TGF-1, IGF-1 levels are elevated,to some extent, it can promote the reproductive function of PCO ratsn.
