摘  要:【摘要】目的评价氟比洛芬酯预先给药对切口痛大鼠中枢β-内啡肽水平的影响。方法SPF级健康雄性SD大鼠54只,6~7周龄,体重180~230g,采用随机数字表法分为3组(n=18):对照组(C组)、切口痛组(I组)和氟比洛芬酯预先给药组(FA组)。切口痛模型制备前30min时I组尾静脉注射脂肪乳1ml,FA组尾静脉注射氟比洛芬酯6mg/kg(用脂肪乳稀释至1m1)。于模型制备前1d(Tn)、模型制备后1、6和12h(T。)时测定机械缩足反应阈(MWT)。于T1-3时痛阈测定后处死大鼠,取脊髓腰膨大和下丘脑弓状核,采用ELISA法测定β-内啡肽含量,采用免疫组化法测定β-内啡肽表达。结果与C组比较,I组和FA组T1-3时MWT降低,I组T2,3时脊髓β-内啡肽含量及表达水平降低,T1时下丘脑弓状核β-内啡肽含量及表达水平升高,FA组T1-3时脊髓和下丘脑弓状核β-内啡肽含量及表达水平升高(P〈0.05);与I组比较,FA组T1-3时MWT升高,脊髓和下丘脑弓状核β-内啡肽含量及表达水平升高(P〈0.05)。结论氟比洛芬酯预先给药对切口痛大鼠产生镇痛效应的机制可能与提高中枢β-内啡肽水平有关。Objective To evaluate the effect of flurbiprofen axetil pretreatment on the level of cen- tral β-endorphin in a rat model of incisional pain. Methods Fifty-four SPF male healthy Sprague-Dawley rats, aged 6-7 weeks, weighing 180-230 g, were divided into 3 groups (n = 18 each) using a random number table: control group (group C) , incisional pain group (group I) and flurbiprofen axetil pretreat- ment group (group FA). At 30 min before the model of incisional pain was established, fat emulsion 1 ml was injected via the caudal vein in group I, and flurbiprofen axetil 6 mg/kg ( diluted to 1 ml in fat emul- sion) was injected via the caudal vein in group FA. The mechanical paw withdrawal threshold (MWT) was measured at 1 day before establishment of the model and 1, 6 and 12 h after establishment of the model (T1-3). The rats were sacrificed after measurement of pain threshold at T1-3, and the lumbar enlargement segment of the spinal cord and hypothalamic arcuate nucleus specimens were obtained for determination of β- endorphin content (by enzyme-linked immunosorbent assay) and β-endorphin expression (by immunohisto- chemistry). Results Compared with group C, the MWT was significantly decreased at T1-3 in I and FA groups, the content and expression of β-endorphin in the spinal cord were significantly decreased at T2,3, and the content and expression of β-endorphin in the hyl)othalamic arcuate nucleus were increased at T1 in group l, and the content and expression of β-endorphin in the spinal cord and hypothalamie areuate nucleus were significantly increased at T1-3 in group FA (P〈0. 05). Compared with group I, the MWT was signifi- cantly increased, and the content and expression of β-endurphin in the spinal cord and hypothalamic areu- ate nuc, leus were increased at T1,3 in group FA (P〈0.05).
