摘 要:目的:研究木瓜苦桑片对高脂高糖联合低剂量链脲佐菌素诱导的Ⅱ型糖尿病大鼠血糖、血脂、肝肾功能和胰腺组织的影响。方法:雄性SD大鼠给予高脂饲料喂养4周后,链脲佐菌素(45 mg·kg^(-1))给药1次,建立糖尿病大鼠模型,将造模成功的大鼠随机分为模型组,二甲双胍组(285 mg·kg^(-1)),木瓜苦桑片高、中、低剂量组(2 600,1 300,650 mg·kg^(-1)),另设正常组,每组10只,每天灌胃给药1次,连续4周,记录每只大鼠24 h的尿量、饮水量、摄食量,分别测定血糖值(FBG),胰岛素(INS),过氧化氢酶(CAT),超氧化物歧化酶(SOD),丙二醛(MDA),一氧化氮(NO),血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白胆固醇(LDL-C),高密度脂蛋白胆固醇(HDL-C),丙氨酸氨基转移酶(AST),天冬氨酸氨基转移酶(ALT),肌酸激酶(CK),肌酐(Cr)和尿素氮(BUN)水平,取胰腺苏木素-伊红(HE)染色观察组织结构。结果:与正常组比较,模型组的大鼠摄食量,饮水量,尿量,FBG,INS,MDA,NO,LDL-C,TC,TG,AST,ALT,BUN,Cr,CK水平均明显升高,CAT,SOD,HDL-C水平降低(P<0.05,P<0.01);与模型组比较,各给药组大鼠摄食量,饮水量,尿量,FBG,INS,MDA,NO,LDL-C,TC,TG,AST,ALT,BUN,Cr,CK水平均明显降低,CAT,SOD,HDLC水平升高(P<0.05,P<0.01);与正常组比较,模型组大鼠胰岛细胞数目减少,排列紊乱,颜色深,细胞间质和细胞间界限不清晰;与模型组比较,各给药组大鼠胰岛细胞数目增多,排列整齐,细胞形态恢复规则。结论:木瓜苦桑片有明显降血糖及血脂作用,可增强机体抗氧化损伤的能力,对肝肾功能具有明显改善作用,可维持胰岛正常结构和功能。Objective:To investigate the effects of Mugua Kusang tablets on blood glucose,lipid,function of liver,kidney and pancreas issues in type 2 diabetes rats induced by high sucrose/high fat diet combined wih low dose streptozotocin(STZ).Method:Diabetes model was induced by feeding with high sucrose/high fat diet for 4 weeks and STZ at dose of 45 mg·kg-1injection in male SD rats,then the successfully modeled rats were divided randomly into the model group,metformin group(285 mg·kg-1),and Mugua Kusang tablets high dose,medium dose and low dose groups(2 600,1 300,650 mg·kg-1).Another 10 normal rats were assigned as the normal control group,n=10 in each group.Intragastric administration was given once a day for 4 weeks,and 24 h urine volume,water intake,and food intake were recorded for all rats.Then the levels of fasting blood glucose(FBG),insulin(INS),catalase(CAT),superoxide dismutase(SOD),methane dicarboxylic aldehyde(MDA),nitric oxide(NO),serum total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),alanine aminotransferase(AST),aspartate transaminase(ALT),creatine kinase(CK),creatinine(Cr) and urea nitrogen(BUN) were measured,and pancreatic structural change was observed by htoxylin-eosin(HE) staining.Result:As compared to the normal control group,the levels of food-intake,water-intake,urine volume,FBG,INS,MDA,NO,LDL-C,TC,TG,AST,ALT,BUN,CK and Cr were increased,while levels of CAT,SOD and HDL-C were decreased in the model group(P〈0.05,P〈0.01).As compared to the model group,the levels of food-intake,water-intake,urine volume,FBG,INS,MDA,NO,LDL-C,TC,TG,AST,ALT,BUN,CK and Cr were decreased,while the levels of CAT,SOD and HDL-C were increased in all treatment groups(P〈0.05,P〈0.01).As compared to the normal control group,islet cells count was decreased,with a disordered arrangement and darker color in model group,and the boundaries were indistinct betw
