摘  要:目的研究氢气(H_2)对于防治氧中毒急性肺损伤(HALI)的作用机制。方法将30只雄性SpragueDawley大鼠随机分为3组:正常对照组、氧中毒模型组(HALI组)和H_2治疗组(H_2组)。其中正常对照组大鼠仅暴露于常压空气,HALI组和H_2组大鼠暴露于100%医用纯氧,每隔12 h分别腹腔注射医用生理盐水和氢饱和生理盐水(10 ml/kg)。60 h后对各组大鼠进行动脉血氧分压检测和肺组织病理学检查,并应用RT-q PCR和Western印迹技术检测肺组织中血红素加氧酶1(human heme oxygenase 1,HO-1)的mRNA转录水平和蛋白表达水平。结果与HALI组相比,H_2组大鼠因慢性纯氧暴露导致的肺损伤程度明显减轻,动脉血氧饱和度升高,HO-1蛋白及其mRNA在肺组织中的表达显著升高。结论 HO-1可能在H_2防治大鼠HALI中发挥重要作用。Objective To investigate the mechanism by which hydrogen( H_2) helps prevent acute lung injury induced by hyperoxia( HALI) in rats. Methods Thirty male Sprague-Dawley rats were randomly divided into three groups: control group,HALI group and H_2 group,with 10 rats in each group. The control group was exposed to air at atmospheric pressure. Rats in HALI and H_2 groups were exposed continuously to pure oxygen( 100% O2) for 60 hours and during this period,10 ml/kg of normal saline or H_2-saturated normal saline was given every 12 hours by intraperitoneal injection to the HALI and H_2 groups, respectively. After treatment, the arterial partial pressure of oxygen was examined and histopathological examination was conducted in each group. Then,RT-q PCR and Western blotting were performed to measure the transcriptional level and protein expression of heme oxygenase 1( human heme oxygenase 1,HO-1) in rat lung tissue. Results Compared with the HALI group,H_2 group showed significantly decreased severity of lung injury and a marked increase in the arterial oxygen saturation. Besides,H_2 treatment induced up-regulation of HO-1 mRNA and protein levels. Conclusion The findings suggest that HO-1 may play an important role in the protection against HALI by H_2.
