摘 要:目的研究防龋用转基因生菜后代植株中外源嵌合基因gtfB-ctxB的遗传稳定性。方法种植T3、T4代外源嵌合基因gtfB-ctxB生菜植株64株、25株,分别在植株幼苗期、开花期、种子成熟期采集叶片、花、茎、种子等不同部位,采用PCR技术检测外源基因gtfB-ctxB,比较T_2~T_4连续世代转基因生菜的阳性率。结果 T_3、T_4代转基因生菜阳性植株分别为40株、15株,PCR阳性率分别为62.5%和60.0%。幼苗期、开花期、种子成熟期植株不同部位(叶片、花、茎、种子)的PCR阳性率检测结果一致。结论嵌合基因gtfB-ctxB能够在防龋用转基因生菜后代植株中稳定遗传。Objective To know genetic stability of anti-caries gene(gtfB-ctxB) in progenies of transgenic lettuce. Methods Genomic DNA was extracted from T_3,T_4 generation of the transgenic lettuce plants in three in different growth stages(young seedling,flowering,seed maturity) and different position(leaves,flowers,stems,seeds). PCR technology was used to detect the target gene gtfB-ctxB in progenies of transgenic lettuce. Identification and selection transgenic lettuce plants which containing the target gene were conducted. The statistics of the positive rate in different generations were collected to compare whether there was a significant differences among the three generations. Results The transgenic lettuce plants genomic DNA of T_3 and T_4 generation which carrying the target gene gtfB-ctxB was amplified by PCR. The positive transgenic plants were 40,15 respectively in each generation. The positive rates were 62.5% and 60.0% respectively. The experimental results of T_3,T_4 generation of the transgenic lettuce plants in three in different growth stages(young seedling,flowering,seed maturity) and different position(leaves,flowers,stems,seeds) were consistent. Conclusion The transgenic lettuce plants carrying the anticaries gene gtfB-ctxB were obtained in the present study and the results demonstrate that the target gene gtfB-ctxB can be stably inherited to the progeny of transgenic lettuce plants.
