摘  要:采用DPPH自由基清除、ABTS自由基清除及FRAP铁离子还原能力三种方法对五株瓦布贝母内生真菌单培养、两两共培养无菌发酵液进行抗氧化活性研究,再测定活性较高组合发酵液的石油醚(30~60℃)、乙酸乙酯和正丁醇提取物的抗氧化活性,以维生素C作为阳性对照。结果发现在瓦布贝母内生真菌两两共培养中,4WBY1+WBS019、6WBY3+WBS019和WBS019+WBS020三个组合无菌发酵液的抗氧化活性至少在两项抗氧化方法评价下均极显著高于单培养。试验还发现,上述三个组合的乙酸乙酯提物和正丁醇提物的抗氧化活性较强;WBS019+WBS020组合的乙酸乙酯提物在1.8 mg/m L时,对DPPH自由基清除率可达74.38%±0.65%;6WBY3+WBS019组合的正丁醇提取物(IC_(50)=0.25 mg/m L)对ABTS自由基清除作用与维生素C(IC_(50)=0.07mg/m L)相对接近;三个组合的各提取物对Fe3+还原能力均较弱,远低于阳性对照。说明在适当条件下开展内生真菌共培养具有潜在利用价值。The purpose of this experiment was to study the antioxidant activities of sterile fermented liquid of single culture and co-culture of five strains of endophytic fungi of Fritillaria unibracteata var. wabuensisby using the method of DP- PH radical scavenging, ABTS radical scavenging and the ability of FRAP iron reduction, and the antioxidant activities of the petroleum ether extract (30-60℃ ), ethyl acetate extract, n-butyl alcohol extract of high active combinations were tested, respectively. At the same time, Vc was used as positive control. The results showed that three co-culture fermented liquid,including 4WBY1 + WBS019,6WBY3 + WBS019 and WBS019 + WBS020, showed significantly higher antioxidant activity than that of single culture in at least two anti-oxidation assays. It was also found that the antioxidant activities of ethyl acetate and n-butanol extracts of previous three combinations were stronger. The DPPH free radical clearance rate of ethyl acetate extract ( 1.8 mg/mL) of WBS019 + WBS020 co-cuhure combination reached to 74.38%±0.65%. The ABTS radical scavenging efficiency of n-butyl alcohol extract (IC50 =0. 25 mg/mL) of 6WBY3 + WBS019 co-cul- ture combination was near to that of vitamin C ( IC50 = 0. 07 mg/mL). The Fe3+ reduction abilities of all extracts of previous three combinations were weak and much lower than that of positive control. These results showed that the endophytic fungi co-culture had potential value under appropriate conditions.
