摘 要:目的探讨碱性成纤维生长因子(bFGF)、血管内皮生长因子(VEGF)在低氧性肺动脉高压(HPH)大鼠肺组织中表达的意义。方法雄性SD大鼠24只,随机分成对照组和实验组,干预后,右心导管法测各组大鼠肺血流动力学指标,称重法测右室肥厚指标(RVHI);光镜观察肺血管病理改变;逆转录聚合酶链反应(RT-PCR)检测bFGFmRNA和VEGFmRNA。结果对照组大鼠的平均肺动脉压(mPAP)和RVHI值显著低于实验组大鼠(P<0.01);光镜显示,实验组大鼠肺血管壁病理学变化较对照组显著。RT-PCR结果提示,实验组大鼠肺组织中bFGF和VEGF值与对照组比较显著升高(P<0.05)。结论低氧可引起bFGF和VEGF的合成增多,bFGF和VEGF参与了低氧性肺动脉高压的发病过程。Objective To explore the expression significance of basic fibroblast growth factor(bFGF) and vascular endothelial growth factor (VEGF)in lung tissue of rats with hypoxic pulmonary hypertension. Methods 24 male SD rats were randomly divided into experimental group and control group,the pulmonary hemodynamic indexes were measured with right heart catheterization method, the right ventrieular hypertrophy indexs(RVHI) were measured by weighing method, the pulmonary artery morphological changes were observed by optical microscope; The expression of bFGFmRNA and VEGFmRNA were detected by reverse transcriptase polymerase chain reaction (RT PCR). Results In the experimental group, mean pulmonary arterial pressure(mPAP)and RVHI were obviously lower than that in the control group (P 〈 0. 01 ) ;Under optical microscopy,the morphological changes of the pulmonary artery wall were more remarkable than that in the control group. The results of RT-PCR showed that the values of bFGF and VEGF were obviously raised in the experimental group than those in the control group(P〈0.05). Conclusion Hypoxia can increase the synthesis of bFGF and VEGF, bFGF and VEGF are involved in the pathogenesis of hypoxic pulmonary hypertension.
