摘 要:目的:探讨白术(Rhizoma atractylodis macrocephalae,RAM)对局灶性脑缺血/再灌注大鼠的神经保护作用。方法:采用Longa等报道的方法,建立局灶性脑缺血/再灌注大鼠模型,通过肌力变化和Longa评分,评价神经功能的恢复情况,同时测定大鼠脑组织超氧化物歧化酶(SOD)、谷胱甘肽(GSH)和过氧化氢酶(CAT)的活性和丙二醛(MDA)含量。结果:(1)行为学评分:神经功能评分:缺血/再灌注组神经功能缺损评分明显高于假手术组(P<0.01),而白术组的神经功能缺损评分明显低于缺血/再灌注组(P<0.01);肌力变化:缺血/再灌注组肌力恢复明显低于假手术组(P<0.01),而白术组的肌力恢复明显高于缺血/再灌注组(P<0.05)。(2)缺血/再灌注组脑组织SOD、GSH和CAT活性显著降低,MDA含量明显增加;而白术组显著增加小鼠脑组织SOD、GSH和CAT活性,降低MDA含量(P<0.01,P<0.05)。结论:白术可以减轻大鼠脑缺血/再灌注,其机制可能与抗氧化有关。Objective: To investigate the effects of rhizoma atractylodis macrocephalae on Neural functional recovery on Cerebral ischemia/reperfusion rat. Methods: Focal cerebral ischemia was induced in anesthetized rats by using Longa re- ported methods model. Neural functional recovery was tested by muscle strength test and Longa's score, and also tested the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH), hydrogen peroxidase(GAT)in brain tissue of Cerebral ischemia/reperfusion rat, and the content of malondialdehyde(MDA)were determined. Results: (1)Behavior- al evaluation: Neural functional: the neural function score in ischemia/reperfusion group were significantly higher than the control group(P^0. 01), and neural function score of RAM group was significantly decreased compared with the ischemia/reperfusion group(P%0. 01) lmuscle strength:the muscle strength recover of RAM group was better than is- chemia/reperfusion group(P〈0. 05). (2)In ischemia/reperfusion group the activities of SOD,GSH,CAT and the con- tent of MDA increase was significantly, rhizoma atractylodis macrocephalae can significantly improve activities of SOD, GSH,CAT and reduce the content of MDA. Conclusion: Rhizome atractylodis macrocephalae could significantly pective the Neural function of Cerebral ischemia/reperfusion rat. The mechanism related to improve activities of SOD, GSH, CAT and reduce the content of MDA in hippocampus.
