摘 要:本实验探讨姜黄素治疗大鼠脊髓损伤后对氧化应激及胶质纤维酸性蛋白(glial fibrillaryacidic protein)表达的影响。将30只大鼠按随机数字表法分为3组:假手术组、模型组、姜黄素(姜黄素组,100 mg/kg),每组10只。假手术组仅咬除椎板,暴露脊髓;模型组、姜黄素组制备脊髓半横断损伤模型。姜黄素组术后即刻按照100 mg/kg剂量腹腔注射姜黄素。模型组腹腔注射等体积DMSO。每日注射1次,连续7 d。假手术组不作处理。在术后当天、7 d、14 d和21 d对3组大鼠行BBB运动功能评分。用化学比色法测定3组术后24 h大鼠血浆中超氧化物歧化酶(superoxide dismutase,SOD)的活力。免疫荧光染色观察GFAP的表达,计算阳性细胞数。结果表明,与模型组比较,术后14 d和21 d,姜黄素组BBB评分显著提高((p<0.05);与模型组比较,术后24 h后姜黄素组SOD活力显著提高(p<0.05);与模型组比较,姜黄素组脊髓损伤部位GFAP阳性细胞数明显减少((p<0.05)。根据本研究结果,我们推断姜黄素在治疗脊髓损伤过程中,可能是通过降低细胞氧化应激水平,进而抑制GFAP表达,发挥对脊髓损伤的治疗作用。The aim of this study was to investigate the changes of oxidative stress and the effect of curcumin on GFAP expression after spinal cord injury. 30 rats were randomly divided into 3 groups, including sham surgery group, model group, curcumin (curcumin group, 100 mg/kg), with 10 rats in each group. In the sham surgery group, the spinal cord was exposed only by removing the neural plate; the model group and the curcumin group were used to establish the hemisection of spinal cord injury model. Curcumin group was intraperitoneally injected with a dose of 100 mg/kg curcumin immediately after the surgery. Control group intraperitoneally received equal volume of DMSO (vehicle). Daily injection once a day lasted for 7 days. The sham surgery group was not treated with any operations. After surgery, BBB motor function scores were respectively evaluated at the surgery's day, 7th day, 14th day and 21st day. The contents of superoxide dismutase (SOD) in plasma of 3 groups were measured by chemical colorimetry at 24 h after operation. The expression of glial fibrillary acidic protein (GFAP) was observed by immunofluorescence staining, and the number of positive cells was counted. The results showed that, the BBB score of curcumin group increased significantly at 14th day and 21st day after operation when compared with themodel group (p 〈0.05). Compared with the model group, the SOD level of curcumin group increased significantly after surgery in 24 h (p〈0.05). Compared with the model group, the number of GFAP-positive cells in curcumin group was significantly decreased (p〈0.05). Based on the results of the study, we concluded that curcumin might play a curative role in the treatment of spinal cord injury by decreasing the level of cells oxidative stress to inhibit GFAP expression.
