山楂叶超高效液相色谱指纹图谱及聚类分析研究

摘　　要：目的:采用超高效液相色谱法(UPLC)结合相似度计算和聚类分析建立山楂叶指纹图谱共有模式,为山楂叶的质量评价提供参考。方法:采用Waters CORTECS UPLC C18色谱柱(3.0mm×100mm,1.6μm),以乙腈-0.1%甲酸水为流动相,梯度洗脱,流速0.2ml/min,检测波长320nm,柱温30℃,进样量3μl。对14批山楂叶进行检测,运用国家药典委员会《中药色谱指纹图谱相似度评价系统2004A版》建立山楂叶UPLC指纹图谱并进行相似度计算和聚类分析。结果:通过聚类分析将14批山楂叶分为Ⅰ、Ⅱ两类,选取第Ⅰ类样品建立山楂叶UPLC指纹图谱共有模式。结论:本研究建立的方法可为山楂叶的质量控制提供参考。Objeclive：To establish hawthorn leaf fingerprint common patterns by using UPLC combined sinailarity calculation and clustering analysis, and provide reference for the quality evaluation of hawthorn leaves. Methods： The UPLC was used with the condition that the column was Waters CORTECS UPLC C18 colullln （3.0mm ×100mm, 1.6μm）; the mobile phase was eluted with gradient by acetonitrilee-0.1% formic acid; the flow rote was 0.2ml/min; the detection wavelength was 320nm; the column temperature was 30~C; the sample volume was 31al. The UPLC fingerprint of 14 batches of hawthorn leaves was established by evaluation system of Waditional Chinese medicine chromatographic fingerprint similarity （2004A version）, and the similarity calculation and cluster analysis were carried out_ Results： The 14 batches of hawthorn leaves were divided into I, II group by cluster analysis; and the hawthom leaf fingerprint common pattem was established based on I group. Conclusions： The method established in this study can provide reference for quality control ofhawthom leaves.
