摘  要:为研究NaAsO_2(亚砷酸钠)中毒对成年SD大鼠纹状体的神经细胞及肝脏形态学的影响,以及NaAsO_2中毒对SD大鼠神经毒性和肝毒性的作用,我们对染毒后的组织进行HE染色并观察。采用12周龄SD大鼠48只,体重200±20 g,随机分组:正常对照组(自由饮水),实验组按剂量递增方式分为低、中、高剂量NaAsO_2染毒组(按照5、10、20 mg/kg的比例配制蒸馏水,自由饮用),各组12只,均采用普通饲料喂养,建造染砷模型12周后取大鼠脑组织纹状体部分,常规石蜡切片后,HE染色,光镜观察大鼠纹状体神经细胞与肝结构的形态学是否发生改变,同时使用试剂盒检测纹状体与肝组织中超氧化物歧化酶(superoxide dismutase,SOD)活力(WST-1法)、还原型谷胱甘肽(reduced glutathione,GSH)(微板法)及丙二醛(malondialdehyde,MDA)(TBA法)的含量。结果显示,利用苏木素-伊红(HE)染色可见:NaAsO_2染毒组的纹状体神经细胞与对照组相比较,神经细胞胞体形态欠规则,数量稀疏且减少,胞浆内可见明显空泡样变,镜下可见坏死水肿的神经细胞。NaAsO_2染毒组的肝组织较正常组出现病理性改变:例如变性、坏死、组织出现水肿及炎性细胞浸润等,与对照组相比,各剂量NaAsO_2染毒组大鼠的纹状体组织及肝脏的SOD活力,GSH含量均降低,差异具有统计学意义(P<0.05);与对照组比较,各剂量NaAsO_2染毒组大鼠的纹状体组织及肝脏的MDA含量均升高,差异具有统计学意义(P<0.05)。由此可知,大鼠纹状体神经细胞形态结构及肝组织结构产生损伤可能由NaAsO_2导致其中毒,同时提示NaAsO_2致大鼠纹状体及肝组织的毒性作用可能与氧化应激相关。To study the effect of NaAsO_2(sodium arsenite) poisoning on the striatum neuronal and liver morphology, and the effect of NaAsO_2 poisoning on neurotoxicity and hepatotoxicity in SD rats. Forty eight 12-week-old SD rats, weighing 200 ±20 g, were randomly divided into normal control group(free drinking water) and experimental group. The experimental group was subdivided into low dose, medium dose and high dose of NaAsO_2 group(according to 5、10、20 mg/kg, in the proportion of preparation of distilled water, free drinking). Each group was fed with ordinary feed. 12 weeks were needed to construct arsenic models. And then, the rat brain tissue parts were took out for HE staining. Light microscopy was used to observe morphological changes of rat striatum nerve cells and liver tissues.The kits were used to detect striatum and liver tissue superoxide dismutase(superoxide dismutase(SOD)(WST-1 method),reduced glutathione(GSH)(Microplate method)and malondialdehyde(MDA)(TBA method).The HE staining showed that the striatum of NaAsO2 group Compared with the control group,the morphology of striatum cells were less regular and the number was sparse and decreased,and the vacuoles were seen in the cytoplasm.The nerve cells of the necrotic edema were observed in the NaAsO_2 group.The pathological changes of the NaAsO_2 group were degeneration,necrosis,tissue edema and inflammatory cell infiltration.Compared with the control group,the content of SOD and GSH in the striatum and liver of the rats exposed to NaAsO_2 decreased obviously(P0.05).Compared with the control group,the content of MDA in the striatum and liver of the rats exposed to NaAsO_2 increased,and the difference was statistically significant(P0.05).The morphological structure of the striatum nerve cells and the damage of the liver tissue structure may be caused by NaAsO_2,and the toxic effects of NaAsO_2 on the striatum and liver tissue of rats may be related to oxidative stress.
