摘  要:目的探讨甲基莲心碱抗光老化的作用机制,为防治皮肤光老化性疾病提供实验依据。方法建立中长波紫外线辐射人皮肤成纤维细胞光老化模型,实验分为对照组、模型组(单纯UVA+UVB照射)和药物组(UVA+UVB照射后加0.4μmol/L,0.8μmol/L,1.6μmol/L甲基莲心碱干预),检测细胞内ROS的水平和SOD和GPx的活力及MDA的含量。结果与模型组相比,甲基莲心碱可以降低ROS水平及MDA含量(P<0.05),同时提高SOD及GPx的活力(P<0.05)。结论适当浓度的甲基莲心碱能降低光老化细胞内ROS引起的氧化损伤,增加细胞内抗氧化酶的活性,减轻细胞膜脂质过氧化作用,延缓了皮肤光老化的进程。Objective To study the anti-photoaging mechanisms of Nef in order to provide a rationale for using Nef in preventing and treating photoaging- associated dermatoses.Methods A photoaging model of human skin fibroblasts was established using UVA and UVB radiation. Fibroblasts were divided into normal control, UVA and UVB-irradiated and treatment groups(UVA and UVB plus Nef0.4μ mol/L,0.8μmol/L,1.6μmol/L). The intracellular levels of ROS and MDA, and activities of SOD and GPx were measured. Results Treatment of fibroblasts with Nef lowered the levels of ROS and MDA while increasing activities of SOD and GPx(P〈0.05). Conclusion Appropriateconcentration of Neferine can decrease oxidative damage, increase activities of antioxidant enzymes,and decrease production of lipid peroxidation in photoaged fibroblasts, leading the delay of skin photoaging.
