针药结合对癫痫大鼠PI3K/AKT信号通路的影响

摘　　要：目的:探讨针灸结合中药对癫痫大鼠PI3K/AKT信号通路的影响。方法:将100只SD雄性大鼠随机分为空白组、模型组、针药组、中药组和西药组5组。空白组、模型组均采用灌服等体积生理盐水;西药组:丙戊酸钠连续灌胃治疗;中药组:祛痰活血方灌胃治疗;针药组:在中药组的基础上,选用百会、神门针刺治疗。结果:行为学结果:与模型组比较,西药组、针药组、中药组大鼠的逃避时间均缩短(P<0.05或0.01),跨越次数均增加(P<0.05或0.01);与西药组比较,针药组逃避时间、跨越次数均无差异,中药组逃避时间、跨越次数均有统计学意义(P<0.05)。PI3K-P85蛋白表达结果:与模型组相比,中药组和针药组均具有统计学意义(P<0.05),西药组具有显著意义(P<0.01);与针药组相比,西药组具有统计学意义(P<0.05)。AKT蛋白表达结果:与模型组相比,中药组和针药组均具有统计学意义(P<0.05),西药组具有显著意义(P<0.01);与针药组相比,西药组具有统计学意义(P<0.05)。结论:针灸结合中药可激活PI3K/AKT信号通路,抑制海马神经细胞的凋亡。Objective：To investigate the effect of combination of acupuncture and medication on signal pathway of PI3K/AKT in epilepsy rats.Methods：80 SD male rats were randomly divided into the blank group（group A）,the model group（group B）,the acupuncture and medication group（group C）,the Chinese medicine group（group D）,and the western medicine group（group E）.Normal saline was orally administrated in group A and group B,group E was given Valproate,group D was applied with Qutan Huoxue formula,GV20 and HT7 were chosen for acupuncture in group C on the treatment basis of group D.Results：In terms of behavior indexes,escape latency was significantly shortened with increased passing times in intervention groups compared to those in group B（P〈0.05,P〈0.01）;in which there was significant difference between group D and group E（P〈0.05）,but there was no significant difference between group C and group E.In terms of PI3K-P85 expression,compared to group B,it was significantly higher in intervention groups（P〈0.05,P〈0.01）,in which there was significant difference between group C and group E.In terms of AKT expression,there was significant difference between group B and other intervention groups（P〈0.05,P〈0.01）,in which the difference between group C and group E was statistically significant（P〈0.05）.Conclusion：Combination of acupuncture and Chinese medication can activate PI3K/AKT signal pathway and inhibit the neurocyte apoptosis in hippocampus.
