摘 要:目的:通过观察卵巢早衰试验大鼠血清中E_2、FSH、β-EP水平和卵巢组织形态的变化,探讨卵巢早衰的病理生理机制以及中药制剂一贯煎对卵巢早衰的作用机制。方法:试验设正常对照组、模型对照组、乙烯雌酚干预组、一贯煎干预组。灌胃给予雷公藤多苷50 mg/kg·d复制卵巢早衰大鼠模型,2周后灌胃给予各组干预药,连续给药6周。之后观察实验大鼠卵巢组织形态学的变化,检测大鼠血清中E_2、FSH、β-EP水平。结果:各组卵巢组织切片形态比较,本实验造模成功,中药制剂一贯煎对卵巢早衰实验大鼠卵巢组织形态有一定的改善作用。与正常对照组比较,模型对照组的E_2、β-EP水平明显降低(P<0.05),FSH的水平显著升高(P<0.05);与模型对照组比较,各干预药组上述指标均有不同程度的改变(P<0.05)。结论:灌胃给予雷公藤多苷可引发大鼠卵巢早衰,中药一贯煎制剂有防治卵巢早衰的作用,其作用机制与能够改变血清中E_2、β-EP、FSH水平有关。Objective:To observe the premature ovarian failure rats’ uterus,ovary,adrenal gland organ index and serum E2,FSH,β-EP levels and explore the pathophysiological mechanism of premature ovarian failure and the action mechanism of Yiguanjian.Methods:The rats were divided into control group,model control group,diethylstilbestrol group,Yiguanjian group.The premature ovarian failure rat model was established by intragastric administration of tripteryium wilfordii polyglycosidium of 50 mg/(kg·d).After 2 weeks of gavage,give the intervention of drug,continuously treating for 6 weeks.The levels of FSH,β-EP and E2 in serum and the organs index of uterus,ovary,adrenal were detected.Results:Compared with the normal control group,the model control group’s E2 and β-EP levels decreased significantly(P〈0.05) and the levels of FSH increased significantly(P〈0.05).Compared with the model control group,the indexes of the intervention groups were improved to different degrees(P〈0.05).Conclusion:Orally given tripterygium wilfordii multi glycosides can cause rats premature ovarian failure and Yiguanjian and its modified formula have the role of prevention and treatment of premature ovarian failure.The mechanism is related to the improvement of E2 and β-EP levels.
