摘 要:目的建立消毒产品中葡萄糖酸氯己定的反相高效液相色谱测定方法。方法样品中葡萄糖酸氯己定经水稀释、过滤,以乙腈-0.02 mol/L磷酸二氢钾(pH=2.5)为流动相,C18色谱柱(4.6 mm×250 mm,5μm)梯度分离,254 nm波长检测,流速为1.0 ml/min,柱温为35℃,外标峰面积标准曲线法定量。结果葡萄糖酸氯己定在0.05 mg/ml^1.00 mg/ml具有良好的线性关系(r=0.999 8),检出限为0.01 mg/ml,回收率为97.4%~100%,RSD为0.50%~1.57%。葡萄糖酸氯己定的分离度符合要求。结论该方法操作简便快速,结果准确可靠,适用于各类不同基质消毒产品中葡萄糖酸氯己定含量的测定。Objective To establish a method for determination of chlorhexidine gluconate( CHG) in disinfection products by reversed phase high performance liquid chromatography. Methods CHG in samples were diluted with water,then filtrated and gradiently separated with mobile phase of acetonitrile-0. 02 mol/L potassium dihydrogen phosphate( pH = 2. 5) by C18column( 4. 6 mm × 250 mm,5 μm),and finally detected at a wavelength of 254 nm. The flow rate was 1. 0 ml/min with the column temperature of 35 ℃. The quantitative analysis of CHG were performed by external standard curve method. Results The method had a good linearility at the range of 0. 05 mg/ml-1. 00 mg/ml( r = 0. 999 8),with the detection limit of 0. 01 mg/ml. The recovery of this method were within 97. 4%-100%,and the RSDs were within 0. 50%-1. 57%. The separation degree of CHG met the requirements. Conclusion The method is simple,rapid,accurate and reliable,which can be used for the determination of chlorhexidine gluconate in disinfection products with different matrix.
