摘  要:目的研究胰岛素对烫伤后未受损皮肤及角质形成细胞、成纤维细胞肿瘤坏死因子-α(TNF-α)和白介素-10(IL-10)基因表达的影响。方法应用20%面积Ⅲ度烫伤大鼠、烧伤血清刺激的皮肤角质形成细胞和成纤维细胞模型,用胰岛素干预后,定量PCR检测大鼠未受损皮肤和模型细胞的TNF-α、IL-10基因表达变化。结果烫伤大鼠及烧伤血清刺激的角质形成细胞、成纤维细胞中TNF-α在6、12、24h明显增高(P<0.05~0.01),而IL-10基因表达无统计学变化,胰岛素干预后,能够明显抑制TNF-α基因而增加IL-10基因表达(P<0.05~0.01)。结论胰岛素可通过对促/抗炎因子基因表达的影响,发挥对皮肤免疫系统的调控作用。Objective To investigate the effect of insulin on gene expressions of TNF-α and IL-10 in uninjured skin after burn,keratinocytes and fibroblasts.Methods SD rats that suffered 20% area III degree burn were used. Burn serum challenged keratinocytes and fibroblasts were pretreated with insulin. Unin- jured skin and cell samples were harvested at the designated time point.The gene expressions of TNF-α and IL-10 were determined by real-time reverse transcription-polymerase chain reaction(RT-PCR).Results The gene expression of TNF-α was elevated significantly in unburned skin, burn serum-challenged keratinocytes and fibroblasts at 6,12 and 24 h, and no statistically significant difference was detected in the IL-10 gene level. Insulin significantly decreased the expression of TNF-α gene but significantly increased the expres- sion of IL-10 gene.Conclusion Insulin might regulate the skin immune response by decreasing the pro-in flammation cytokine and increasing the anti-inflammatory cytokine after burn injury.
