摘 要:目的建立一种同时分离和测定大花罗布麻叶中6种黄酮类成分的微乳液相色谱方法。方法用HPLC法测定,色谱柱为Zorbax SB-C_(18)(4.6mm×150mm,5μm);流速:0.8ml·min^(-1);检测波长:360nm;柱温:30℃;采用经过优化后的微乳体系为流动相,其组成为:2.0%Genapol X-080-3.0%正丁醇-0.6%乙酸乙酯-0.2%三乙胺-94.2%水溶液(20mmol·L^(-1) H_3PO_4调节至pH 5.0)。结果大花罗布麻叶中6种黄酮类成分在15min内达到基线分离,在1~500mg·L^(-1)范围内,6个黄酮成分的线性相关系数≥0.9998,回收率98.9%~100.8%。结论本微乳液相色谱法可应用于大花罗布麻叶中6种黄酮成分的同时分离和测定。Objective To establish a new method for simultaneous separation and determination of six flavonoids in Poacynum hendersonni (Hook. F) Woodson leaf extract using reversed phase high perform ance liquid chromatography with a microemulsion system mixture as the mobile phase.Methods Separation was performed on the Zorbax Extend-Qs column at a UV detection wavelength of 360 nm. The flow rate was 0.8 ml ·min ^-1.The optimized microemulsion mobile phase consisted of 2.00/00 ( V/V ) of Genapol X- 080, 3.0% (V/V) n-butanol, 0.6% (V/V) ethyl acetate, 0.3 % (V/V) triethylamine and 94.2% ( W/V ) of an aqueous solution,while pH was adjusted to 5.0 with 20 mmol·L^-1 phosphoric acid.Results The calibration curve for six flavonoids was linear in the range of 1--500 mg·L^-1 with a correlation coeffi- cient greater than 0.9998.The mean recovery ranged from 98.9~ to 100.8~.Conclusion The microemulsion liquid chromatography method can be used to separate and determine six [avonoids in Poacynum hendersonni (Hook.F) Woodson leaf extract.
