HPLC-ELSD法测定复方天麻蜜环糖肽片中黄芪甲苷含量

摘　　要：目的建立HPLC-ELSD法测定复方天麻蜜环糖肽片中黄芪甲苷的含量。方法色谱柱为Agilent C_(18)柱(250mm×4.6mm,5μm);流动相:乙腈-水(32∶68);流速:1.0ml·min^(-1);柱温:30℃;ELSD参数:漂移管温度为45℃;氮气吹扫流速为450kPa。采用外标二点法对数方程计算含量。结果黄芪甲苷进样质量浓度线性范围为0.0253~0.5060mg·ml^(-1),线性回归方程为log y=1.596log x+6.695,r=0.999(n=6);平均回收率为98.99%,RSD为1.87%(n=6)。结论该方法简便、准确、专属性强,可用于测定复方天麻蜜环糖肽片黄芪中黄芪甲苷的含量。Objective To determine astragaloside IV in compound armillaria mellea polysaccharide and poly peptide tablets.Methods HPLC-ELSD was adopteed. The chromatographic column was AgilentC18 （250 mm × 4.6 mm,5 μm）.The mobile phase consisted of acetonitrile-water（32 ： 68） at a flow rate of 1.0 ml·min^-1 .The column temperature was kept at 30 ℃.The drift tube temperature of ELSD was 45 ℃ and the nitrogen flow-rate was 450 kPa.Two-point external standard method of logarithmic equation was used to calculate the content of astragaloside IV.Results A good linearity was observed within the range of 0.0253--0.5060 mg ·ml ^-1.The regression equation was logy =1.596logx ＋6.695（ r =0.999）,the average recovery was 98.99%, and the RSD was 1.87%（ n =6）. Conclusion This method is simple,precise and reproducible,which can be used for the quantitative control of corn pound armillaria mellea polysaccharide and polypeptide tablets.
