摘 要:目的:通过构建2型糖尿病(T2DM)大鼠妊娠模型,探讨左归复方对2型糖尿病妊娠大鼠模型胰腺组织CD4、TGF-β1表达的影响。方法:将40只雌性SD大鼠随机分为5组,其中4组以高糖高脂饮食并腹腔注射链脲佐菌素(STZ)建立2型糖尿病大鼠模型组,采用电化学法对各组大鼠按照血糖水平排序,随机将40只雌性SD大鼠分为正常组、中药低剂量组、中药高剂量组、达美康组、模型组,2∶1雌雄比例合笼,建立2型糖尿病大鼠妊娠模型。观察大鼠一般情况,妊娠第0.5天开始对以上5组分别予等容量0.9%氯化钠注射液、低剂量左归复方、高剂量左归复方、达美康混悬液、蒸馏水灌胃21d,观察各组大鼠胰腺组织病理形态学的变化,免疫组化法检测胰腺CD4、TGF-β1蛋白的表达水平。结果:与正常对照组比较,模型组胰腺CD4水平显著升高,TGF-β1水平显著降低,差异均有统计学意义(P<0.01或P<0.05);与模型组比较,中药低、高剂量组胰腺CD4水平显著降低,达美康组TGFβ1水平显著升高,差异有统计学意义(P<0.01或P<0.05);中药低剂量组较达美康组CD4、TGFβ1水平低,差异有统计学意义(P<0.01)。结论:左归复方能够改善2型糖尿病大鼠CD4、TGF-β1蛋白的表达。Objective: To investigate the effect of compound Zuogui prescription on the expression of CD4 and transforming growth factor-beta 1( TGF-β1) in pancreatic tissue in pregnant rats with type 2 diabetes mellitus( T2DM) by establishing a rat model of pregnancy and T2DM. Methods: A total of 40 female Sprague-Dawley rats were randomly divided into five groups. The rats in four groups were given high-glucose and high-fat diet and intraperitoneal injection of streptozotocin to establish a rat model of T2DM. The electrochemical method was used to measure blood glucose,and the rats in each group were placed in an order of blood glucose level. The 40 rats were randomly divided into normal control group,low-and high-dose traditional Chinese medicine( TCM) group,Diamicron group,and model group and mated with male rats at a ratio of 2∶ 1 to establish a rat model of pregnancy and T2DM. The rats' general status was observed,and since 0. 5 day of pregnancy,the rats in the above five groups were given equal volumes of 0. 9% sodium chloride injection,low-dose compound Zuogui prescription,high-dose compound Zuogui prescription,Diamicron suspension,and distilled water by gavage for 21 days. The morphopathological changes of the pancreas were observed and immunohistochemistry was used to measure the protein expression of CD4 and TGF-β1. Results: Compared with the normal control group,the model group had a significant increase in CD4 and a significant reduction in TGF-β1 in the pancreas( P〈0. 01 or P〈0. 05). Compared with the model group,the low-and high-dose TCM groups had a signif icant reduction in CD4,and the Diamicron group had a significant increase in TGF-β1( P〈0. 01 or P〈0. 05). The low-dose TCM group had significantly lower levels of CD4 and TGF-β1 than the Diamicron group( P〈0. 01). Conclusion: Compound Zuogui prescription can improve the expression of CD4 and TGF-β1 in rats with T2DM.
