摘 要:【目的】魔芋软腐病是魔芋生产中的一个重要细菌性病害,严重威胁着魔芋产量。【方法】本实验拟利用血清学技术建立快速检测魔芋细菌性软腐病菌的方法。【结果】采用棋盘滴定法确定抗原和抗血清的最适工作浓度分别为106CFU/m L和1∶10000;病原菌检测灵敏度为103CFU/m L;抗血清与其它细菌标准菌株的交叉反应结果均呈阴性,结果表明该方法具有较高的特异性。将该方法检测了24份魔芋种球、病土和带病植株,检出率高达91.67%。【结论】该技术可以有效的检测魔芋软腐病原菌胡萝卜软腐果胶杆菌(Pectobacterium carotovora subsp.carotovora,p.c.c.),为生产上魔芋种球快速检测软腐病菌提供了方法。[ Objective] The soft rot of Amorphophallus konjac(A, konjac) is the major disease affecting the yield of A. konjac. [ Method ] In this study, a method for rapid detection of the soft rot pathogen of Amorphophallus konjac was established by serological technique. [ Result ] Using checkerboard titration, the best working concentration of antigen and antiserum were 106 CFU/mL and 1 : 10000 respeefively. The sen- sitivity of the serum was tested, and the lowest p. c. c. suspension was 103 CFU/mL. Cross reactions of antisera with other bacteria were de- tected, all results were negative. The cross assay indicated that this method had high serological specificity. The method has been used to de- tect 24 samples, with the detective rate 91.67 %. [ Conclusion]The results indicated that this method can be used to detect the pathogens of p. c. c.. This is very useful for diagnosing latent infection ofp. c. c of seed tubers in the early stage.
