摘  要:【目的】为探明荔波瑶山鸡POU1F1基因多态性与繁殖性状的关系,为荔波瑶山鸡的选择性育种提供参考。【方法】针对POU1F1基因设计6对引物,采用PCR测序法筛选出多态位点,并利用荔波瑶山鸡母系60个个体进行繁殖性状的关联分析。【结果】荔波瑶山鸡POU1F1基因有8个多态位点,其中POU1F1基因58(A→G)与300日龄产蛋量、首次就巢日龄、300日龄就巢次数均呈显著或极显著相关(P<0.05或P<0.01);65(T→C)与300日龄产蛋量、首次就巢日龄、300日龄就巢次数呈显著相关(P<0.05);11041(T→C)与300日龄产蛋量、首次就巢持续时间、首次就巢至重新开产间隔均呈显著或极显著相关(P<0.05或P<0.01)。【结论】POU1F1基因58(A→G)和65(T→C)可作为300日龄产蛋量、首次就巢日龄和300日龄就巢次数的辅助选择标记;11041(T→C)可作为300日龄产蛋量、首次就巢持续时间、首次就巢至重新开产间隔的辅助选择标记。[ Objective] The stydy aims to analyze the associations of gene POUIF1 with chicken egg laying traits and provide references for selection breeding of Libo Yaoshan chicken. [ Method] Six primers were designed and eight polymorphic sites were selected, and the association analysis was performed in 60 chickens of Libo Yaoshan female line by PCR-sequencing methods. [ Result ] 58 (A-→G) mutation sites were significantly associated with the total egg number from 120 to 300 d of age (EN300), age at first broodiness behavior (AFBB) and times of broodiness behavior from 120 to 300 d of age (TBB300) (P 〈 0.05 or P 〈 0. 01 ). 65 (T→C) mutation sites were significantly associated with EN300, AFBB and TBB300 ( P 〈 0.05 ). 11041 (T→C) mutation sites were significantly associated with EN300. Duration of age at first broodiness behavior (DAFBB) and interval of age at first broodiness behavior between anew laying (IAFBBA) ( P 〈 0.05 or P 〈 0. 01 ). [ Conclusion ] 58 (A→G) and 65 (T→C) may be regarded as an effective molecular marker on the MAS( Marker Assisted Selection) program for EN300, AFBB and TBB300; 11041 (T→C) may be regarded as an effective molecular marker for EN300, DAFBB and IAFB-A.
