摘  要:分别用细粒棘球绦虫原头蚴(PSC)、包囊壁(HC)和包囊液(HF)刺激培养在24孔板上的小鼠巨噬细胞RAW 264.7,并用PBS作为对照,用G riess试剂检测不同时间点一氧化氮(NO)的浓度。结果显示,在短时期(48 h)内,PSC刺激巨噬细胞后在第8、24和48小时均产生大量NO,与PBS对照组相比均差异显著(P<0.05);HC也可刺激巨噬细胞产生NO,与PBS对照组相比,在刺激后第24和48小时也均差异显著(P<0.05);HF在刺激后第4、24和48小时均产生少量NO,但与PBS对照组相比均差异不显著(P>0.05)。结果表明,巨噬细胞可通过不断释放大量NO来抵御原头蚴的侵染,包囊对宿主具有损害作用,同时又能通过降低NO的产生来减弱宿主对原头蚴的杀伤作用,包囊液对巨噬细胞释放NO的作用不显著。本研究为进一步揭示宿主在抵抗寄生虫感染时NO的作用机制奠定了基础。Murine macrophages RAW264.7 were cultured on 24-well plates,and were stimulated with the protoscoleces(PSC),hydatid cyst wall(HC) and hydatid cyst fluid(HF) of Echinococcus granulosus.Meanwhile,the PBS group was designed as negative control.The concentration of nitric oxide(NO)was detected by Griess reagent at different time points.The results showed that NO was increased at 8hours,24 hours and 48 hours post-stimulation(PC) in PSC group,respectively,and there were significant differences compared with that of the PBS control group(P〈0.05).HC also stimulated the NO production by induced macrophages,and the results showed that there were significantly differences compared with that of the PBS control group at 24 hours and 48 hours PC(P〈0.05).HF induced a small quantity of NO at4 hours,24 hours and 48 hours PC but there were not significantly differences compared with that of the PBS control group(P〈0.05).The results indicated that the macrophages could resist the infection of protoscolices by producing NO.The hydatid cyst wall has a damaging effect on the host,but also can weaken the harmful effects to the protoscolex by reducing the production of NO.The effect of hydatid cyst fluid was not significant to NO production by macrophages.The experiment will be useful for further study on the mechanism of NO against parasite infection.
