摘 要:目的利用高能超声破碎法和超滤膜获得黄芩中较高纯度的总黄酮成分。方法以黄芩中黄芩苷化合物为对照品,采用紫外-可见分光光度法进行含量测定。对比超声、微波、回流和高能超声破碎提取法,并利用正交试验,以时间、乙醇浓度、料液比和次数4个因素进行考察,采用Biomax-5膜进行超滤膜分离。结果单因素试验中以高能超声破碎提取法提取含量较高。正交试验结果:时间5 min,乙醇浓度为50%,料液比1∶10,提取3次。超滤膜分离条件:以流量为1.5 L/min、压力为0.8 kg进行膜分离。结论高能超声破碎法和超滤膜的结合,用于黄芩中黄酮类化合物的纯化,方法可行,产品纯度高,方法具有良好的应用前景。Objective To obtain high purity flavonoids ingredients of Scutellaria baicalensis Georgi using the method of ultrasonic emulsification and membrane separation. Methods The content of Baicalin in Scutellaria ba- icalensis was determined by UV visible spectrophotometry. Compare ultrasound, microwave,reflux and ultrasonic emul- sification method ; and orthogonal experiment was used to investigate the 4 factors : ultrasonic emulsification time, etha- nol concentration, solid-liquid ratio and times. Biomax-5 membrane was used to make ultraflltration membrane separa- tion. Results In the single factor experiment, the extraction rate of ultrasonic emulsification extraction was higher. Or- thogonal test results showed the ultrasonic emulsification time was 5 min, ethanol concentration was 50% , the solid liq- uid ratio : 1 : 20, extraction : 3 times. Membrane separation conditions : the membrane separation was done at a flow rate of 1.5 L/min and ultrafiltration membrane pressure of 0. 8 kg. Conclusion The combination of ultrasonic emulsification and membrane can be used for the purification of flavonoids in Scutellaria baicalensis. The method is feasible, the purity of the product is high, and the method has a good application prospect.
