摘  要:目的探讨miR-132表达上调对局灶性脑缺血大鼠皮层血管生成素1(Ang^-1)及其内皮特异性酪氨酸激酶受体(Tie2)表达的影响。方法40只成年健康雄性SD大鼠按随机数字表法分为假手术组、脑缺血组、miR-132模拟物组和阴性对照组,每组10只。后3组按照改良Longa线栓法制备成大脑中动脉闭塞模型,其中miR-132模拟物组和阴性对照组于脑缺血5min时分别将miR-132模拟物15μg和阴性对照物15μg经侧脑室注入。缺血后24h取脑组织,采用2,3,5.氯化三苯基四氮唑(TTC)染色计算各组大鼠脑梗死体积,采用实时荧光定量PCR检测缺血侧皮层组织miR-132、Ang^-1、Tie2 mRNA表达,采用Western blotting检测缺血侧皮层组织Ang^-1、Tie2、CD31、血管内皮生长因子(VEGF)蛋白表达。结果miR-132模拟物组脑梗死体积[(27.92±3.05)mm^3]显著低于脑缺血组和阴性对照组[(51.34±2.86)mm^3和(50.46±2.571mm^3],差异均有统计学意义(P〈0.05)。miR-132模拟物组缺血侧皮层组织miR-132、Ang—1、Tie2mRNA相对表达量及Ang^-1、Tie2、CD31、VEGF蛋白相对表达量均明显高于阴性对照组、脑缺血组和假手术组,且阴性对照组和脑缺血组均明显高于假手术组,差异均有统计学意义(P〈0.05)。结论上调miR-132表达可改善脑缺血大鼠脑组织缺血性损伤,其机制可能与促进Ang^-1/Tie2表达进而促进缺血脑组织血管再生有关.Objective To investigate the effect of up-regulation of miR-132 on expressions of angiopoietin-1 (Ang^-1)/endothelium-specific tyrosine kinase receptor 2 (Tie2) in focal cerebral ischemia reperfusion injury of rats. Methods Forty adult healthy SD rats were randomly divided into sham-operated group, cerebral ischemia group, miR-132 mimic group and negative control group (n=10). The models of middle cerebral artery occlusion in the later three groups were established by using modified Longa suture method. Rats in the miR-132 mimic group and negative control group were injected miR-132 mimic 15 tzg and negative control 15 p^g via paracele. Rats in each group were sacrificed 24 h after ischemia, and the brain tissues were collected; the total infarct volumes were calculated by TTC staining. The mRNA expressions ofmiR-132, Ang^-1 and Tie2 in ischemic cerebral cortex tissues were detected by real-time fluorescence quantitative PCR. The protein expressions of Ang^-1, Tie2, CD31 and vessel endothelial growth factor (VEGF) in ischemic cerebral cortex tissues were detected by Western blotting. Results The total infarct volume in the miR-132 mimic group was (27.92±3.05) mm^3, which was significantly smaller than that in the cerebral ischemia group and negative control group ([51.34±2.86] mm^3 and [50.46±2.57] mm^3, P〈0.05). The relative mRNA expression levels of miR-132, A rig^-1, Tie2, and the protein expression levels of Ang^-1, Tie2, CD31, VEGF in the ischemiccerebral cortex tissues of the miR-132 mimic group were significantly higher than those in the negative control group, cerebral ischemia group and sham-operated group (P〈0.05); and those in the negative control group and cerebral ischemia group were significantly higher than those in the sham-operated group (P〈0.05).
