摘 要:目的观察礞石滚痰汤对戊四氮(PTZ)致痫小鼠惊厥行为和额叶皮层电活动的改变。方法腹腔注射PTZ 45mg/(kg·d)建立癫痫动物模型,将制备成功的小鼠随机分为4组:模型组(B)、礞石滚痰汤低(C)、高(D)剂量组、丙戊酸钠(VPA)组(E),每组各10只,另设空白对照组(A)10只。模型组给予生理盐水灌胃,C组、D组、E组分别给予礞石滚痰汤低、高剂量及丙戊酸钠等不同因素灌胃处理。给药10d后,除空白对照组外,各组小鼠再次腹腔注射PTZ 45mg/(kg·d),观察动物行为的改变,同时利用脑立体定位仪记录各组癫痫小鼠脑部单侧额叶皮质的脑电变化。结果行为学观察显示,与模型组比较,药物治疗组小鼠癫痫发作的潜伏期延长(P<0.05),1h强直-阵挛发作的次数减少(P<0.05),发作持续时间缩短(P<0.05)。脑电图结果显示,与模型组比较,药物治疗组能有效抑制小鼠痫性放电。结论礞石滚痰汤能有效抑制戊四氮致痫小鼠惊厥行为发作和额叶皮层的异常放电。Objective To investigate the effect of Mengshi Guntan Tang on the acute seizure model and electroencephalogram of the model. Methods Mice acute seizure model of epilepsy were made by PTZ with intraperitoneal injection,Except for blank controlgroup( A),40 refractory epilepsy rats were randomly divided into four groups:,model group( B),low( C),high( D) dose of Mengshi Guntan Tang groups,and VPA group( E). The same amount of physiologicalsaline was given to model group,while other groups were given low,high dose of Mengshi Guntan Tang and VPA,respectively. Except blank control,other rats were ip.PTZ45mg/( kg·d) again after they were treated for 10 days. The changes of unilateral frontal cortex electroencephalogram( EEG)in mice were recorded by brain solid positioner and their ecliptic behaviors were observed. Results(1)Showed by animals’ behaviors,in treatment groups,the latent period of paroxysm was extended significantly( P 〈 0. 05),the number of titanic-clonic seizures was decreased obviously during 1 h( P 〈 0. 05),and the mean duration of seizures was reduced compared with epileptic control( P 〈 0. 05).(2)Showed by EEG,compared with model group,treatment groups can restrain brain electrical abnormal discharge of mice. Conclusion Mengshi Guntan Tang can significantly restrain the acute epileptic seizure and decrease abnormal epileptic wave of the frontal cortex of mice.
