摘 要:以梅花鹿茸角为研究对象,探讨胰岛素样生长因子1(insulin-like growth factor-1,IGF-1)对鹿茸软骨细胞Ⅰ型胶原(collagenⅠ,ColⅠ)表达的影响。分离培养鹿茸软骨细胞,分别添加IGF-1重组蛋白、IGF-1受体抑制剂(PQ401)和IGF-1重组蛋白,同时构建IGF-1过表达载体并合成siRNA,转染鹿茸软骨细胞,通过荧光定量PCR方法检测软骨细胞去分化标志分子ColⅠ表达的变化。荧光定量PCR结果显示,添加IGF-1重组蛋白后,随着时间的增加,ColⅠmRNA在鹿茸软骨细胞中的表达量逐渐下降,添加PQ401后,IGF-1抑制ColⅠmRNA表达的作用减弱;转染IGF-1过表达载体后,ColⅠmRNA在鹿茸软骨细胞中的表达量显著降低,而转染IGF-1siRNA后其表达量则显著升高。结果表明,IGF-1可能在鹿茸软骨细胞去分化过程中起重要的作用。This study aimed to use the sika deer as a model to study the influence of IGF-1 on the expression of Col I in antler chondrocytes. The chondrocytes were separated from sika deer antlers,cultured and were treated with recombinant human IGF-1 protein (rIGF-1),both rIGF-1 and PQ401 ,and transfected with IGF-1 over-expression plasmid or IGF-1 siRNA, respectively. The expression of Col I which, a well-known marker for chondrocytes dedifferentiation, was detected by real-time PCR. The results showed that administration of rlGF-1 to antler chondroctyes resulted in an obvious decrease of Col I mRNA levels, while PQ401 pretreatment could dramatically attenuate the effects of rIGF-1 on the expression of Col I mRNA. After transfection with IGF-1 over-ex- pression plasmid, the expression of Col I mRNA was obviously reduced in antler chondrocytes compared with control. Conversely,knockdown IGF1 with specific siRNA could increase the ex- pression of Col I in antler chondrocytes. These results indicate that IGF-1 may play an important role in process of antler chondrocyte dedifferentiation.
