脆性三联组氨酸蛋白和B细胞淋巴瘤-2蛋白在胃癌组织中的表达及其与幽门螺旋杆菌感染的相关性研究

摘　　要：目的观察脆性三联组氨酸(FHIT)蛋白和B细胞淋巴瘤-2(Bcl-2)蛋白在胃癌组织中的表达及其与幽门螺旋杆菌感染的相关性研究。方法选取36例胃癌手术的癌组织、17例癌旁组织和17例慢性萎缩性胃炎组织为研究对象。用免疫组织化学法检测组织中FHIT蛋白以及Bcl-2蛋白的表达情况。用甲苯胺蓝法和14C呼气试验检测幽门螺旋杆菌的感染情况。结果癌组织、癌旁组织和胃炎组织的FHIT蛋白阳性表达率分别为44.44%,76.47%和70.59%,Bcl-2蛋白阳性表达率分别为63.89%,29.41%和35.29%,癌组织的上述指标与胃炎组织和癌旁组织比较,差异均有统计学意义(均P<0.05)。FHIT蛋白和Bcl-2蛋白在胃癌组织表达呈负相关(P<0.001)。幽门螺旋杆菌阳性组织和阴性组织的FHIT蛋白阳性表达率分别为57.69%和10.00%,Bcl-2蛋白阳性表达率分别为42.31%和80.00%,差异均有统计意义(均P<0.05)。结论胃癌组织中FHIT蛋白和Bcl-2蛋白表达呈负相关,幽门螺旋杆菌感染对胃癌组织中FHIT蛋白和Bcl-2蛋白表达造成影响,三者均于胃癌发生发展过程中起重要作用。Objective To explore the expressions of fragile histidine triad protein（ FHIT） protein and B cell lymphoma-2（ Bcl-2） protein,and the clinical relationships between their expression and Helicobacter pylori infection in gastric carcinoma. Methods A total of 36 cases of gastric cancer tissues,17 cases of adjacent tissues and gastritis tissues were selected. The expressions of FHIT protein and Bcl-2 protein were detected by immunohistochemistry SP method,and the Helicobacter pylori infection was detected by the toluidine blue method and14 C breath test.Results The positive-expression rates of FHIT protein in gastric cancer,adjacent non-cancerous tissues and gastritis tissues were44. 44%,76. 47%,70. 59%. The positive-expression rates of Bcl-2protein in gastric cancer,adjacent non-cancerous tissues and gastritis tissues were 63. 89%, 29. 41%, 35. 29%. There were significant differences between the cancer tissues and adjacent non-cancerous tissues and gastritis tissues（ all P〈0. 05）. The expressions of FHIT protein and Bcl-2 protein in gastric carcinoma were negatively correlated（ P〈0. 001）. The positive-expression rates of FHIT protein inHelicobacter pylori positive and negative tissues were 57. 69% and 10. 00% respectively,and Bcl-2 protein were42. 31% and 80. 00% respectively, and the differences were statistically significant（ all P〈0. 05）.Conclusion Expressions of FHIT protein and Bcl-2 protein are negatively related in gastric carcinoma, and Helicobacter pylori infection may influence the expressions of FHIT protein and Bcl-2 protein in gastric carcinoma,and they play an important role in the occurrence and development of gastric cancer.
