摘 要:目的:正交设计法优化喜树碱胶束的制备工艺。方法:采用透析法制备喜树碱胶束,在单因素考察的基础上,以投料比、p H值、透析温度为影响因素,药物包封率为指标,应用正交设计法优化载药胶束的制备工艺,并对最佳工艺进行重复性实验。结果:喜树碱胶束的最佳制备工艺为:聚合物材料与药物的投料比为1:10、p H值为5、透析温度为30℃。重复性实验结果表明,载药胶束的平均包封率为51.86%,RSD为0.11%。结论:最佳制备工艺制得胶束的包封率均高于正交试验结果,该制备工艺稳定可行,成功实现了难溶性药物喜树碱的增溶,为喜树碱胶束的制备提供了实验依据,为提高此类难溶性药物的临床疗效奠定了基础。Objective:The research aimed to optimize the preparation technology of camptothecin micelles by orthogonal design.Methods:The drug loaded micelles were prepared by dialysis method.The orthogonal tests were used to optimize the preparation technology involving feed ratio,pH value and temperature.Results:The optimal preparation technology of dialysis method was that the feeding ration was 1:10,pH value was 5,the temperature was 30℃.The repeat test shows that the encapsulation rate was up to51.86%with high stability as RSD=0.11%.Conclusion:The optimized preparation technology of camptothecin micelles was stable and feasible.Successfully solve the problem of insoluble drug CPT.
